摘  要:目的:探讨地塞米松及1α-25-(OH)2D3诱导的致耐受树突状细胞(TolDC)治疗免疫性血小板减少症小鼠的疗效。方法:用地塞米松及1α-25-(OH)2D3诱导TolDC,光镜观察形态特点,流式检测细胞膜CD80、CD86水平。将TolDC腹腔注射给免疫性血小板减少症小鼠,观察症状并检测小鼠的血小板水平及血小板相关自身抗体。结果:地塞米松及1α-25-(OH)2D3诱导的树突状细胞毛刺少、突起短,CD80、CD86水平低,符合TolDC的特点。TolDC治疗组小鼠血小板水平较模型组高(P<0.05),血小板相关自身抗体水平较模型组低(P<0.05)。结论:地塞米松及1α-25-(OH)2D3诱导的TolDC能避免免疫性血小板减少症小鼠的血小板下降,血小板相关自身抗体减少,为其应用于自身免疫性血小板减少症的治疗提供了理论依据。Objective To explore the effect of dexamethasone and 1α-25-(OH)2-D3 -induced dendritic cells on immune thrombocytopenia in mice. Methods Bone marrow-derived dendritic cells were cultured with dexamethasone and 1α-25-( OH )2-D3. Morphology was observed, and levels of menbrane CD80, CD86 were detected by FACS. Platelets were counted, and levels of platelet related autoimmune antibodies were detected by FACS in immune thrombocytopenia mice treated with tolerogenic dendritic cells or saline. Results Dexamethasone and 1α- 25-(OH)2-D3-induced tolerogenic dendritic cells had less and shorter dendrites and levels of CD80 and CD86 were lower compared with mature dendritic cells. Platelets counts were increased and level of platelets related autoimmune antibodies were decreased in immune thrombocytopenia mice treated with tolerogenic dendritic cells compared with those treated with saline. Conclusion Dendritic cells treated by dexamethasone and 1α-25-(OH)2-D3 have protective effect on immune thrombocytopenia mice, which povides an evidence for further research in the effect of tolerogenic dendritic cells on immune thrombocytopenia purpura.
