凋膜止崩液宫腔靶向给药对ADUB模型大鼠子宫内膜组织中MMP-1、TIMP-1、ER及亚型表达的影响

摘　　要：目的探讨凋膜止崩液宫腔靶向给药治疗无排卵性功血(ADUB)子宫内膜增生症的可能机制,为临床治疗ADUB提供一种新方法。方法卵巢去势后利用雌激素负荷法建立ADUB子宫内膜增生症大鼠模型,凋膜止崩液宫腔内给药后处死,留取子宫标本,采用RT-PCR检测法测定各组大鼠子宫内膜组织中基质金属蛋白酶-1(MMP-1)和基质金属蛋白酶抑制因子-1(TIMP-1)基因表达水平的变化;采用免疫组化技术检测各组大鼠子宫内膜组织中雌激素受体(estro-gen receptor,ER)、ERα、β亚型的表达变化。结果模型对照组与空白组比较,MMP-1mRNA表达明显升高,TIMP-1mRNA表达降低,差异有统计学意义(P<0.05)。实验大、小剂量组与模型对照组比较,MMP-1mRNA表达降低,TIMP-1mRNA表达升高,差异有统计学意义(P<0.05),而大小剂量组间比较无显著性差异(P>0.05)。模型对照组与空白组比较,ER、ERα、β表达明显升高,差异有统计学意义(P<0.05)。实验大、小剂量组与模型对照组比较,ER、ERα、β表达下调,差异有统计学意义(P<0.05),而实验大、小剂量组间比较无显著性差异(P>0.05)。结论凋膜止崩液宫腔靶向给药后ADUB模型大鼠子宫内膜组织中MMP-1mRNA表达下降,TIMP-1mRNA表达升高,ER、ERα、β表达下调,可能是其治疗子宫内膜增生症的机制。Objective To explore the mechanism of Diaomozhibeng liquid in treatment in endometrium hyperplasia （EH） and provide to a new method of clinically treating endometrial hyperplasia. Methods Rat model was induced by ovariectomy combining with estrogen. The expression levels of MMP - 1, TIMP - 1, ER,ERα, ERβ, MMP - 1, TIMP - 1 were checked by real - time PCR,and ER,ERα, ERβwere checked by immunohistoehemistry. Results Compared with the control group, the expression of MMP - lmRNA in model group increased, while that of TIMP - lmRNA decreased. The difference was statistically significant （ P 〈 0.05 ）. Compared with model group, the high dose of Diaomozhibeng liquid and low dose of Diaomozhibeng liquid ,the expres- sion of MMP - lmRNA decreased and the expression of TIMP - lmRNA increased. The difference was statistically significant （ P 〈 0.05 ）. While compared with the high dose of Diaomozhiheng liquid and low dose of Diaomozhibeng liquid, there was signifi- cant difference （P 〉 0.05 ）. Compared with the control group, the expression of ER, ERα, ERβ in model group significantly increased （ P 〈 0.05 ）. Compared with model group, the high dose of Diaomozhibeng liquid and low dose of Diaomozhibeng liquid, the ex- pression of ER,ERα、β decreased. The difference was statistically significant （P 〈 0.05 ）. While compared with the high dose of Diaomozhibeng liquid and low dose of Diaomozhibeng liquid, there was no significant differentce （ P 〉 0.05 ）. Conclusion The mechanism of treating endometrial hyperplasia with Diaomozhibeng liquid uterine cavity target intervention may be associated with reducing the expression of MMP- 1mRNA ;ER,ERα、βand increasing the expression of TIMP- 1mRNA.
