热敏灸对抑郁症模型大鼠内分泌激素的调节作用

摘　　要：目的制备抑郁症大鼠模型,研究热敏灸对抑郁症模型大鼠血清内分泌激素水平的影响,阐明热敏灸治疗抑郁症的机理。方法采用雌性SD大鼠,联合孤养和慢性不可预见性温和应激(CUMS)制备抑郁症大鼠模型。用Open-Field测试大鼠的直立活动和水平活动得分;1h内大鼠摄入饮用水和蔗糖水量计算糖水消耗百分比;大鼠阴道细胞涂片观察大鼠动情周期规律;放射免疫分析法(RIA)测定大鼠血清雌二醇(E2)、甲状腺素(T4)水平,酶联免疫吸附分析法(ELISA)测定大鼠血清皮质酮水平。结果 21d CUMS可使大鼠的水平活动得分和直立活动得分减少、糖水消耗百分比降低,动情周期紊乱,血清E2水平降低、皮质酮水平增高,但T4水平无明显变化;而热敏灸具有提高大鼠的水平活动得分和直立活动得分、增加大鼠糖水消耗百分比,调整动情周期以及提高血清E2水平和降低血清皮质酮水平。结论热敏灸可能通过调整体内内分泌激素水平而达到改善抑郁症。Objective To observe the effect of heat - sensitive moxibustion on serum hormone level in depression model rats. Methods Female SD rats were induced into depression model by giving alone in combination with 21 days of chronic unexpected mild stress. Open -field was used to test crossing and rearing. Sucrose consumption test was used to calculate the percentage of sucrose intake. Estradiol and thyroxine were measured by radioimmunoassay （RIA） , corticosterone was measured with enzyme - linked immunosorbent assay（ ELISA ） . Results Compared with rats in the model group, both crossing scores and rearing scores were increased, the percentage of sucrose intake was increased, serum estradiol level was upgraded while corticosterone level was deduced in the heat - sensitive moxibustion group rats. Conclusion Heat - sensitive moxibustion can improve the symptoms of de- pression, which might be through regulating the level of hormones.
