摘  要:目的研究维生素E对阿霉素引起的心肌细胞过氧化损伤的保护作用。方法以原代培养的乳鼠心肌细胞建立损伤模型。在实验中,将细胞分为3组:空白对照组、模型组(2mg/L)的阿霉素与乳鼠心肌细胞共培养。药物修复组:在加入阿霉素的同时,加入维生素E(150、300、600μg/ml与心肌细胞共同培养。测定各组心肌细胞的乳酸脱氢酶I(LDHI)含量、丙二醛(MDA)含量及一氧化氮合酶(NOS)活性的变化。结果模型组心肌细胞的LDHI含量增高(P<0.01),MDA含量增加(P<0.05),NOS活性增高(P<0.01).而药物修复组使心肌细胞的LDHI含量降低(P<0.01),MDA含量降低(P<0.05),NOS活性降低(P<0.01),且降低的幅度随药物浓度的增加而增加。结论维生素E和对受损的心肌细胞有明显的保护作用。Objective To investigate protective effects of vitamin E on myocardium injury caused by free radicals. Methods Myocardial cells of new-born rats were incubated with ADR(2 mg/L)to induce oxygen free radical injury in model group;same dose of ADR and three different dose of VE were administered in therapeutic group. Content of LDHI,content of MDA, activity of NOS were measured. Results Content of LDHI: compared with that of control group,the content of ADR was increased in model group(P〈0.01). Compared with that of model group, the content of LDHI was decreased in VE group and Se group(P〈0.01). Content of MDA:compared with that of control group,the content of ADR was increased in model group (P〈0.05). Compared with that of model group, the content of MDA was decreased in VE group. Activity of NOS: compared with that of control group, the content of ADR was increased in model group(P〈0.01). Compared with that of model group, the activity of NOS was decreased in VE group (P〈 0.01). Conclusion VE has greatly protective effects 0n free radical injury induced by ADR.
