摘 要:目的研究甘草次酸对大鼠心室肌细胞钠离子通道电流(INa)的影响,探讨甘草次酸在离子通道水平抗心律失常的作用机制。方法急性分离雄性SD大鼠心室肌细胞,通过膜片钳全细胞记录技术观察甘草次酸对大鼠心室肌细胞INa的影响。结果甘草次酸可剂量依赖地抑制大鼠心室肌细胞钠电流。1、5、10μmol/L甘草次酸使大鼠心室肌细胞钠电流自[(-4.26±0.15)nA],分别降低至[(-3.54±0.1)nA],[(-2.19±0.09)nA],[(-1.25±0.08)nA],差异均有统计学意义。1、5、10μmol/L甘草次酸对大鼠心室肌细胞钠电流峰值的抑制率分别为(16.08±2.3)%、(50.82±3.56)%、(75.98±5.12)%。10μmol/L甘草次酸使INa电流-电压关系曲线上移,但不改变其激活电位和峰电位。结论甘草次酸可浓度依赖地抑制大鼠心室肌细胞INa,其抗心律失常作用可能与此相关。Objective To study the effects of glycyrretinic acid (GA) on the sodium ion channel currents (Ina) of rats' ventricular myocardial cells, and to explore its anti-arrhythmic mechanisms at the ion channel level. Methods Single ventricular myocardial cells was isolated from SD rats. The whole cell patch clamp was used to record the effects of GA on I,a of rats' ventricular myocardial cells. Results GA could inhibit I,, of rats' ventricular myocardial cells dose-dependently. GA at I, 5, and 10 ~mol/L decreased I,a of rats' ventricular myocardial cells from ( -4. 26 +0. 15) nA to ( -3.54 +0. 10) nA, ( -2.19 ~0.09) nA, and ( -1.25 ~0. 08) nA, respectively. GA at 1,5, and 10 ijmol/L inhibited INa by 16.08% ~2.3%, 50.82% ~3.56%, and 75. 98% ~5. 12%, showing statistical difference when compared with the control group (P 〈0.05). GA at 10 ~mol/L shifted IN. current-volt- age curve more positively, but the activation potential and the peak potential were not changed. Conclusion GA inhibited the INa of rats' ventricular myocardial cells dose-dependently, which was possibly associated with its anti- arrhvthmia effects.
