摘 要:抗体库为抗体工程带来了革命性的突破,其核心是将抗体的基因型与表型耦联,从抗体库中找到针对特异抗原的抗体基因。但是抗体工程发展所面临的两大难点是如何构建大库容量的抗体库和提高抗体的亲和力。抗体库方法和技术的出现使得改造工程抗体更方便、更有效。利用不同抗体库方法构建大库容量的突变抗体库,然后通过抗体库技术在体外模拟抗体亲和力成熟过程获得高亲和力的抗体。本文简要综述了抗体库方法和技术的最新进展。Antibody library has brought a revolutionary breakthrough in the field of antibody engineering. The core of antibody library depends on coupling genotype with phenotype, which provides an effective way to get the gene sequence of antibodies against specific antigen from antibody library. While constructing antibody library with large capacity and improving antibody affinity are two major difficulty of the development of antibody engineering. The emergence of antibody library methods and technologies makes engineering antibodies renovation easier and more effective. By adopting different antibody library methods, mutant antibody library with larger capacity have been constructed. Then the high-affinity antibody has achieved through antibody library technologies by imitating the maturation process of the antibody affinity in vitro. In this review, the advances of antibody library methods and technologies are summarized briefly.
