茎段部位和去叶处理对亮叶忍冬试管苗生长的影响

摘　　要：本文旨在研究接种茎段的部位及去叶处理对亮叶忍冬试管苗生长的影响。在添加了30 g/L蔗糖的1/4 MS培养基中,分别接种带有茎尖的上部单节茎段和不带茎尖的中下部单节茎段,以及去双叶、去单叶和不去叶的中下部单节茎段进行培养。结果表明,带有茎尖的上部茎段的萌芽率和繁殖系数明显高于不带茎尖的中下部茎段,但两者的生根率、根长和根数等指标无明显差异;去双叶茎段的萌芽率和繁殖系数显著高于不去叶茎段,但其生根率、根长则明显低于不去叶茎段。在采用单节扦插法进行亮叶忍冬试管苗茎芽增殖培养时,应去除中下部茎段的全部两个叶片,在生根培养中则应保留其叶片。This article aimed at studying the effects of stem section with different position and defoliation treatment on the growth ofLonicera nitida ＇Maigrun＇ plantlet in vitro. The top stem section with stem apex, the middle-basic stem section and the middle-basic stern section with different leaf cutting treatments （2 leaves cut down, 1 leaf cut down or no leaf cut down） were respectively cultured in 1/4 MS medium with 30 g/L sugar added. The results show that both the germination rate and the propagation coefficient of the top stem section with stem apex were obviously higher than that of the middle-basic stem section, while there were no obvious difference in the rooting rate, the root length and the root number between the top stem section and the middle-basic stem section. Both the germination rate and the propagation coefficient of the middle-basic stem section with 2 leaves cut down were obviously higher than that of the stem section with no leaf cut down, but the rooting rate, the root length of the stem section with 2 leaves cut down was obviously lower than that of the stem section with no leaf cut down. During the Lonicera nitida ＇Maigrun＇ micro-shoot multiplication culture by one-nod stem section cutting, the all 2 leaves of the middle-basic stem section should be cut down, but in rooting culture the leaves should not be cut down.
