摘  要:目的:探讨高温环境下运动至力竭对大鼠肾脏水通道蛋白2(AQP2)的影响。方法:32只健康雄性Wistar大鼠随机分为正常对照组(C)、常温力竭组(E)、高温暴露组(H)和高温力竭组(HE),每组8只。常温环境25℃,湿度50%;高温环境38℃,湿度50%。C组不进行任何处理,E组在常温环境下以20 m/min的速度运动到力竭,H组在高温环境下暴露1小时,HE组在高温环境下以20 m/min的速度运动到力竭,记录E、HE组大鼠运动到力竭的时间。实验前各组大鼠称量体重,E、HE组大鼠力竭后即刻称量体重,H组暴露1小时后称量体重;麻醉后取血和肾脏,测试血浆渗透压、血清抗利尿激素(AVP)浓度,RT-PCR法测定大鼠肾脏抗利尿激素V2受体(AVPV2R)和AQP2mRNA表达,Western blot法测定肾脏AVPV2R蛋白表达,免疫荧光检测磷酸化的AQP2蛋白表达。结果:(1)E组大鼠力竭运动时间为(163.00±26.04)min,HE组大鼠力竭运动时间(28.00±11.44 min)明显缩短(P<0.01)。(2)HE组大鼠体重丢失%明显低于E组(P<0.05);E组和HE组大鼠血浆渗透压和血清AVP浓度明显高于C组(P<0.05)。(3)E组、H组和HE组大鼠肾脏AVPV2R mRNA和AQP2 mRNA表达上调,显著高于C组(P<0.01),AVPV2R蛋白表达无明显变化;E组、H组和HE组大鼠磷酸化的AQP2表达增强。结论:高温环境下大鼠运动能力降低,机体脱水明显,AVP起到了调节维持体液平衡的作用。高温环境下力竭运动后AQP2在转录水平发生改变,磷酸化的AQP2表达增强。Objective The purpose of this study was to investigate the change of aquaporin-2(AQP2) in the kidney after exercise to exhaustion in a high healthy Wistar rats were randomly divided into four normal environment group (E), heat stress group (H) temperature environment. Methods Thirty two male groups :control group (C),exhaustive exercise in a and exhaustive exercise in a high temperature environment group (HE). Each group contains 8 rats. The temperature in normal environment was set at 25℃ ,and the humidity at 50%. The high temperature environment was created in a chamber with 38℃temperature and 50% humidity. Group C received no any treatment, group E exercised in a normal environment at the speed of 20 m/min, group H was exposed in the high temperature environment for 1 hour, and group HE exercised in the high temperature environment at the speed of 20m/min. The exhaustive exercise duration of group E and HE were measured. Rats in each group were weighted before the ex- periment. The weight of groups E and HE was obtained immediately after exhaustive exercise,and of group H 1 hour after exposing to heat. The blood,hypothalamus and kidney were sampled. The concentration of plasma osmolality,serum AVP,AVPV2R mRNA and protein expressions,and AQP2 mRNA and AQP2 phosphorylated protein expressions in each group were measured. Results (1)The exhaustive exercise duration was 163.00±26.04 min and 28.00±11.44 min,respectively,for group E and group HE, which were significantly shorter than the group H. (2)The weight loss of group HE was significantly lower than group E (P 〈 0.05) ;plasma osmolality and serum AVP concentration of group E and HE were significantly higher than group C(P 〈 0.05). (3)The expression of AVPV2R mRNA and AQP2 mRNA of group E, H,and HE were significantly higher than group C (P 〈 0.01 );AVPV2R protein expression remained unchanged ;the phosphorylated AQP2 expression of group E, H, and HE enhanced.
