脐带间充质干细胞染色体核型制备及安全评估

摘　　要：目的探讨脐带间充质干细胞染色体核型制备方法,评估脐带间充质干细胞安全性。方法对培养至第2、5、8、10、11代的共15例脐带间充质干细胞进行染色体制备,检查其有无异常。结果成功制备染色体标本,成功率为100%,正常核型率为100%。结论该脐带间充质干细胞染色体制备方法实验结果稳定、成功率高。脐带间充质干细胞在培养至11代时未见异常染色体核型。Objective To investigate methods for preparing chromosome karyotype of umbilical mesenchymal stem cells,and to evaluate the safeness.Methods The chromosomes of umbilical mesenchymal stem cell cultured for 2,5,8,10,11 generations were prepared and analyzed.Results The chromosome karyotype of umbilical mesenchymal stem cell were prepared successfully,with a success rate of 100%.The rate of normal karyotype was 100%.Conclusion The preparation method is effective and stable,with high success rate.No abnormality was found until the 11th generation.
