摘 要:目的通过检测miR-155和miR-146a在系统性红斑狼疮(SLE)患者外周血单个核细胞(PBMC)和血浆中的表达,探讨miR-155和miR-146a的表达在SLE发病中的作用。方法 SLE患者20例,健康对照组15例。取外周血分离PBMC和血浆,提取和纯化miRNA,实时定量PCR检测miR-155和miR-146a的表达,以小分子RNAU6作为内对照,计算标准化后的2-△Ct值表示miRNA的相对表达含量。结果 SLE患者PBMC中miR-155和miR-146a的表达水平是健康对照组的3.07倍(P<0.01)和1.91倍(P=0.056);SLE患者血浆中miR-155和miR-146a的表达水平比对照组低,但差异无统计学意义;同时SLE患者中蛋白尿阳性组血浆中两种miRNAs表达水平有降低的趋势,活动组患者血浆中两种miRNAs表达水平也有降低的趋势,但差异无统计学意义。结论 SLE患者PBMC中miR-155和miR-146a异常高表达,但血浆miR-155和miR-146a的表达水平有下降的趋势。Objective To investigate the expression of miR-155 and miR-146a in peripheral blood mononuclear cells (PBMC) and plasma of patients with systemic lupus erythematosus (SLE). Methods PBMC and plasma were separated from the peripheral blood of 20 patients with SLE (the study group) and 15 healthy individuals (the control). Total RNAs were isolated and miRNAs were purified. The levels of miR-155 and miR-146a were determined by quantitative reverse transcription PCR (qRT-PCR), using U6 as housekeeping control. The amount of target miRNA was normalized relative to the amount of U6 (ΔCt =ΔCtmiRNA-ΔCtU6). Relative expression levels were expressed as 2-ΔCt. Results The expression of PBMC miR-155 was significantly higher in the study group than that in the control group (P〈0.05); the expression of PBMC miR-146a was higher in the study group than that in the control group, but no statistically significant difference was observed (P=0.056). The expression of plasma miR-155 and miR-146a in the study group was lower than that in the control group, with no statistically significant difference (P〉0.05). The expression of plasma miR-155 and miR-146a were respectively lower but not significantly in proteinuria positive group and active group than those in proteinuria negative group and stable group (P〉0.05). Conclusion The expression of PBMC miR-155 was higher in SLE patients, but the expression of plasma miR-155 and miR-146a decreased in SLE patients.
