摘  要:目的研究低频超声联合盐酸氨溴索对表皮葡萄球菌(S.epidermidis)RP62A生物被膜(biofilm,BF)结构的影响。方法建立成熟RP62A BF模型。分组处理,XTT法测BF细菌活性变化;激光共聚焦显微镜结合ISA软件定量分析BF;扫描电镜观察BF结构改变。结果单独低频超声作用BF无效果,1.875 mg/mL浓度氨溴索能有效清除BF,较空白对照组区域孔率增大,平均扩散距离和结构熵减小,P<0.05;氨溴索联合低频超声(1、0.5 h)组效果更加显著,与单独氨溴索组相比,P<0.05;扫描电镜显示低频超声能明显促进氨溴索对BF的作用,使BF变薄,结构趋于单一疏松。结论低频超声能显著促进氨溴索对表皮葡萄球菌BF的清除作用。Objective To study the effect of low-frequency ultrasound combined with ambroxol hydrochloride on structure of S. epidermidis RP62A strain's biofilm (BF). Methods Mature biofilm model of S. epidermidis RP62A strain was established, after divided different groups with different interventions, the metabolic activity of the bacteria was quantified by methyl tetrazolium salt (XTT) reduction assay; the BF structures were evaluated by eonfocal laser scanning microscope (CLSM) with image structure analyzer(ISA) software; Changes in the structures of BF were observed by scanning electron microscope(SEM). Results Low frequency ultrasound alone showed no effect on BF; Ambroxol with a concerntration of 1. 875 mg/mL effectly destroyed BF, areal porosity (AP) in- creased,average diffusion distance (ADD) and textual entropy (TE) decreased, compared with the control groups, P〈0. 05;Ambroxol combined with Low-frequency ultrasound(1,0.5 h) showed more obviously effect than the Am- broxol group,P〈0.05 ; SEM showed Low-frequency ultrasound increased the role of Amhroxol on BF, the BF got thinner, more simple and loose. Conclusions Low-frequency ultrasound combined with ambroxol showed an en- hanced bactericidal activity on RP62A BF.
