摘　　要：目的:探讨镉(Cd)引起卵泡细胞凋亡与分拣蛋白(Sort1)表达的关系。方法:利用流式细胞仪检测大鼠卵泡颗粒细胞的凋亡;利用免疫组织化学技术检测大鼠卵巢中Sort1的表达部位,在此基础上利用实时荧光定量PCR技术和蛋白免疫印迹技术检测CdCl2处理后大鼠卵泡中Sort1表达量的变化。结果:5μmol/L、20μmol/L和50μmol/L 3个浓度的CdCl2染毒处理均可引起大鼠卵泡颗粒细胞的凋亡;Sort1主要表达于大鼠卵巢颗粒细胞和卵泡膜细胞中;3个浓度的CdCl2均可使大鼠卵巢中Sort1的表达量显著增加(P<0.05)。结论:Sort1可能参与Cd诱导的卵泡颗粒细胞的凋亡过程。Objective： To study the relationship between the expression of Sortl and the apoptosis of granulosa cells in ovary. Methods： The localization of sortl in rat ovary was investigated by immunohistochemistry （IHC）, and then real-time PCR and Western blotting were employed to determine expression levels of sort1 in rat ovary after the treatment with differnt concentrations （0 mmol/L, 5 mmol/L, 20 mmol/L, 50 mmol/L） of CdC12. Results： CdC12 with different concentrations （5 mmol/L, 20 mmol/L, 50 mmol/L） could all induce the apoptosis of granulosa in rat follicles. IHC staining showed that sort1 protein was localized mainly in granulosa and theca cells of follicles. The expression of Sortl in rat ovary was increased significantly after treated with different concentrations of CdC12 （P〈0.05）. Conclusion： Sortl may be an important factor involving in the process of apoptosis induced by cadmium.
