摘　　要：目的探讨妊娠糖尿病(GDM)患者血清及胎盘中内脂素(visfatin)的表达水平。方法采用酶联免疫吸附法及放免法分别测定30例妊娠糖尿病(GDM组)及30例正常孕妇(对照组)血清内脂素及胰岛素水平,计算胰岛素抵抗指数(HOMA-IR)及体质量指数(BMI)。采用逆转录-聚合酶链反应(RT-PCR)及蛋白印迹法,检测两组胎盘内脂素基因mRNA的表达及蛋白的表达水平。结果①GDM组血清visfatin、FBG、FIN、HOMA-IR、BMI水平显著高于对照组(P<0.05)。②GDM组胎盘组织中visfatin mRNA及蛋白表达水平显著高于对照组(P<0.05)。③血清内脂素水平与HOMA-IR呈正相关(P<0.05)。结论 GDM孕妇血清及胎盘内脂素水平的升高可能参与了GDM的发生发展,血清内脂素与胰岛素抵抗相关。Objective To investigate the expression level of visfatin in placenta and serum of gestational diabetes mellitus（GDM）.Methods The levels of serum visfatin,fasting blood glucose（FBG）and fasting insulin（FIN）were measured.The insulin resistance index（HOMA-IR）was calculated.The expression of visfatin mRNA and protein were separately determined by reverse transcription polymerase chain reaction（RT-PCR）and western-blot from 30 pregnant women with GDM（GDM group）and 30 pregnant women with normal glucose（control group）.Results ①The serum levels of visfatin,FBG,FIN,BMI and HOMA-IR were significantly higher in GDM group than those in the control group（P0.05）.②The levels of visfatin mRNA and visfatin protein expression in placenta tissue in GDM group were significant higher than those in the control group（P0.05）.③ There was positive correlation between serum visfatin level and HOMA-IR.Conclusion The increased visfatin in serum and placenta maybe involved in the development of GDM.
