葡萄糖转运蛋白介导孕酮对于氧糖剥夺PC12细胞的防护作用

摘  要:目的探讨葡萄糖转运蛋白抑制剂细胞松弛素B(CB)对孕酮抗氧糖剥夺(OGD)防护PC12细胞的作用及机制。方法选取生长状态良好及诱导分化均一的PC12细胞随机分为5组:正常(normal)组、氧糖剥夺(OGD)组、孕酮(PROG)组、孕酮+细胞松弛素B(PROG+CB)组、细胞松弛素B(CB)组,后4组均行氧糖剥夺。XTT检测细胞活力,胎盘蓝染色法计数活细胞,氧化酶法测定培养基中糖含量。上述检测在OGD后24h进行。结果 OGD后PC12细胞肿胀变圆,突起减少或消失,细胞活力(P<0.01)和活细胞数(P<0.05)降低,培养液葡萄糖含量明显升高(P<0.05)。PROG减轻胞体水肿,增加细胞活力(P<0.01)和活细胞数(P<0.05),降低培养液葡萄糖含量(P<0.05)。先用CB抑制葡萄糖转运蛋白,再给PROG(PROG+CB组)则抵消PROG的防护作用。单用CB(CB组)则细胞活力仅为1.8%,活细胞数为2.9%,培养液中葡萄糖含量明显升高。可见,孕酮对氧糖剥夺PC12细胞防护作用可被葡萄糖转运蛋白的抑制剂抵消。结论葡萄糖转运蛋白可能是介导孕酮防护氧糖剥夺PC12细胞的重要分子靶标。Objective To observe the effects of cytoehalasin B on progesterone(PROG) protection against oxygen- glucose deprivation(OGD) injury in pheoehromoeytoma cell line (PC12) cells and to investigate the action mechanism of PROG. Methods PC12 cells which grew well and differentiated uniformly were randomly divided into five groups : normal group that PC12 cells were cultured in medium contained oxygen and glucose as usual. In other four groups the cells were subjected to OGD-injury including OGD model, progesterone(PROG) , PROG + eytoeha]asin B (CB) , and CB group. Cell vi- ability was detected by (XTT) ,living cells was counted by using Trypan Blue staining, and suger content in medium was measured with glucose oxidase assay. Above tests were made at the time point of 24h after OGD. Results OGD made the cell body swell and round, the neurite shorter or even disappeared, the cell vitality (P 〈 0.01 ) and the living cells (P 〈 0.05 ) sharply dropping, while suger content in medium increasd. PROG made the cell body edema alleviating, the cell vital- ity ( P 〈 0. O1 ) and the living cells ( P 〈 O. 05 ) going up, the suger content in medium going down ( P 〈 0.05 ). The cell pro- tection of PROG was abolished if PC12 cells were firstly treated with eytochalasin B( in PROG + CB group ). The cell vitali- ty was 1.8% and the living cells was 2.9% only when glucose transporters were inhibited with eytoehalasin B ,cell damage was more badly. From the above data,it implied that the protection of PROG on PC12 cells injuried by OGD could be coun- teracted by eytoehalasin B, a glucose transporter (GLUT) inhibitor. Conclusion Glucose transporter (GLUT) may be a key molecular target in PROG protecting PC12 cells against OGD-injury.
