摘 要:目的:研究健脾复方对人大肠癌移植瘤裸小鼠肿瘤环氧化酶2(COX-2)、核因子κB(NF-κB)和激活物蛋白1(AP-1)表达的影响。方法:将40只近交系(BALB/C)裸小鼠建立人大肠癌原位移植瘤模型,随机分入健脾复方组、塞来昔布组、5-Fu组、模型对照组,另设空白对照组,共分组治疗8周。采用逆转录聚合酶链反应(RT-PCR)方法检测COX-2 mRNA表达,采用免疫组化法检测AP-1和NF-κB的表达。结果:健脾复方组、塞来昔布组和5-Fu组裸小鼠COX-2表达均显著低于模型对照组,且与空白对照组相似(P=0.0019)。健脾复方组、塞来昔布组、5-Fu组和模型对照组AP-1表达高于空白对照组(P=0.0065)。健脾复方组和空白对照组NF-κB表达低于5-Fu组和模型对照组(P<0.05),且健脾复方组与空白对照组间差异未见显著性。结论:健脾复方可以下调COX-2的表达,其机制可能与下调其上游因子NF-κB有关。健脾复方对AP-1表达的影响尚需进一步研究。Objective: To investigate the effect of Jianpi Compound Recipe on COX-2,AP-1,and NF-κB expression in nude mice with human colorectal cancer grafts.Method: Human colorectal cancer orthotopic transplanted tumor model was established in 40 BALB/C nude mice.These nude mice were randomized into Jianpi Compound Recipe group,celecoxib group,5-Fu group,and model control group.A blank control group was also established.All groups received respective treatments for 8 weeks.The expression of COX-2 mRNA was assessed by RT-PCR,the expression of AP-1 and NF-κB were assessed by immunohistochemistry method.Results: COX-2 mRNA expressions were significantly lower in Jianpi Compound Recipe group,5-Fu group,Celecoxib group compared with model control group(P=0.0019),and were similar with blank control group.Expressions of AP-1 were significant higher in Jianpi Compound Recipe group,5-Fu group,Celecoxib group and model control group compared with blank control group(P=0.0065).Expressions of NF-κB was significantly lower in Jianpi Compound Recipe group and blank control group compared with 5-Fu group and model control group(P〈0.05),there was no significant difference between Jianpi Compound Recipe group and blank control group.Conclusion: Jianpi Compound Recipe could down-regulate the expression of COX-2,this may due to the down-regulation of NF-κB.The effect on AP-1 needs further investigation.
