摘  要:目的:建立高效液相-三重串联四极杆质谱联用测定昆明山海棠提取物中雷公藤甲素含量的方法。方法:以乙腈-水(0.05%甲酸)为流动相,0.8mL/min,柱温30℃,Aglient C18柱分离,采用大气压化学电离源(APCI),正离子检测,选择离子监测模式(SIM)。结果:雷公藤甲素在40.72~1628.8ng/mL范围内线性关系良好,平均加样回收率为92.5%,最低定量限27.15ng/mL。结论:该方法选择性强,灵敏度高,可为昆明山海棠提取物中雷公藤甲素含量测定提供参考。Objective:To establish a method for determination of triptolide by high-performance liquid chromatography-tandem mass spectrometry in extracts of Tripterygium hypoglacum ( Levi. ) Hutch. Methods : Triptolide was separated on a Aglient C18 column using acetonitrile and water ( 0.05%formic acid ) as mobile phase at the flow of 0.6mL/min and column temperature at 30~C . MS determination was performed by Atmospheric Pressure Chemical Ionization ( APCI ) in positive mode using Selected Ion Monitoring (SIM)mode. Result: The triptolide was linear within the range of 40.72-1628.8ng/mL ( r=0.9996 ), and the average ( n=6 ) recovery was 98.8% with the RSD of 9.6%. Conclusions : The method is selective, sensitive and applicable to the quantitative determination of triptolide in' extracts of Tripterygium hypoglacum.
