摘 要:为了分析大鼠FSH分泌受体后信号转导机制。将GTH细胞用PMA或H7处理后,用GnRH脉冲刺激,再用ELISA法检测其FSH分泌量,并与空白对照组比较。结果表明:PMA能显著提高GTH细胞中PKC活性,H7能显著降低GTH细胞中PKC活性,PMA或H7都不会影响GTH细胞的cAMP含量,GTH细胞PKC活性显著影响FSH的分泌,FSH随着PKC活性的升高而升高,随着cAMP的降低而降低。得出PKC-Ca2+是GnRH脉冲刺激所引起的FSH分泌的受体后的信号转导途径。In order to analyze the post-receptor signal transduction mechanism of the secretion of FSH in rats. The GTH cells was stimulated by GnRH after the activity of PKC in them was treated by PMA or H7. FSH was detected by ELISA, which was compared with the control group. The results showed that PMA could significantly improved the activity of PKC in GTH cells, which H7 could significantly reduced. Neither PMA nor H7 could not influenced the content of cAMP significantly in GTH ceils. The activity of PKC in GTH ceils influenced the secretion of FSH significantly, that was to say, the level of FSH rose with the rise of PMA and fallen with the falling of H7. The conclusion was drawn that PKC-Ca2 + was the post-receptor signal transduction pathway that the ceils stimulated by GnRH impulse secreted FSH.
