摘 要:目的:探讨自体骨髓间充质干细胞(BMSCs)移植及联合丹参注射液对于兔肢体缺血模型的疗效。方法:新西兰兔18只经丝线结扎血管造成肢体缺血模型,随即平均分为3组:对照组即生理盐水组(NS);单纯移植组即单纯自体骨髓间充质干细胞移植组(BMSCs);联合用药组即自体骨髓间充质干细胞移植联合丹参注射液组(BMSCs+D)。建立模型1周后,单纯移植组、联合用药组取自体培养BMSCs,在腓肠肌部位皮下多点注射,对照组予以生理盐水,联合用药组在单纯移植组的基础上合并丹参注射液耳静滴。常规饲养2周后,(1)超声检测血管生成指标:股浅动脉内径、血流信号、峰值血流速度、加速时间;(2)免疫组化染色观察比较各组抗Brdu染色阳性细胞;(3)HE染色观察比较各组血管生成情况。结果:(1)超声结果:①2D超声显示联合用药组与单纯移植组股动脉及股浅动脉近段管径较对照组明显增宽(P<0.05),且联合用药组较单纯移植组增宽明显(P<0.05);②CDFI超声显示联合用药组与单纯移植组较对照组股动脉血流信号明显增强,且联合用药组较单纯移植组血流信号增强明显;③彩色多普勒图像显示单纯移植组和联合用药组股动脉内血流的峰值流速较对照组明显增高,血流加速时间明显小于对照组(P<0.05),且联合用药组略优于单纯移植组(P>0.05)。(2)免疫组化染色:单纯移植组和联合用药组均检测到抗Brdu染色阳性细胞,且联合用药组多于单纯移植组。(3)HE染色:单纯移植组和联合用药组组织血管密度明显高于对照组,且联合用药组高于单纯移植组。结论:联合用药组与单纯移植组在反映血管生成相关方面明显优于对照组,且联合用药组较单纯移植组为优。Objective: To investigate the effect of autologus bone marrow mesenchymal stem cell(BMSCs) transplan- tation and its combination with Danshen injection for the New Zealand rabbit limb ischemia model. Methods: 18 New Zealand rabbits, through the thread ligation of vessels resulting in limb ischemia models, were divided into 3 groups: the control group, namely the physiological saline group (NS); the BMSCs transplantation group, the autologous bone marrow mesenchymal stem cell transplantation group (BMSC); the combined treatment group, the autologous bone marrow mes- enchymal stem cell transplantation combined with Salvia miliorrhiza injection group (BMSCs+D). A week after the model was established, in the BMSCs transplantation group and the combination group, autologous BMSCs cultured were injected into the gastrocnemius muscle subcutaneously at muhipoints, the control group was treated with saline and the combination group was treated with auricular intravenous drip of Danshen on the basis of BMSCs transplantation. After two weeks of conventional breeding, (1) ultrasonic tests were conducted of angiogenesis indicators, superficial femoral artery diameters, blood flow signals, peak velocity and acceleration time, (2) by immunohistochemical staining, Brdu staining positive cells were observed and compared in all the groups and (3) through HE staining angiogenesis was observed and compared. Re- suits:(1) Ultrasound findings: (1)2D ultrasound showed, in the combination group and the BMSCs transplantation group, femoral artery and proximal superficial femoral artery diameters widened significantly compared with the control group (P〈 0.05), and in the combination therapy group, the diameters widened more significantly than in the BMSCs transplantation group (P〈0.05); (~) CDFI ultrasound showed, in the combination group and the BMSCs transplantation group, the femoral artery blood flow signals were enhanced more obviously than in the control group and the s
