摘 要:目的探讨Notch-1信号通路对胰腺癌BxPC3细胞增殖和凋亡的影响。方法体外培养胰腺癌BxPC3细胞,用小干扰RNA(small interference RNA,siRNA)的方法抑制Notch-1的表达,Western印迹方法检测Notch-1、Bcl-2及Bcl-xL蛋白表达情况;MTT法检测细胞增殖;ELISA法检测细胞凋亡。结果抑制Notch-1表达能明显抑制胰腺癌BxPC3细胞生长(P<0.01),显著下调Bcl-2和Bcl-xL水平,并使BxPC3细胞凋亡显著增多(P<0.01)。结论 Notch-1信号通路促进胰腺癌BxPC3细胞增殖,抑制胰腺癌细胞凋亡。Objective To observe the effect of Notch-1 signaling pathway on cell growth and apoptosis in BxPC3 pancreatic cancer cells. Methods Human pancreatic cancer cell line BxPC3 was cultured. Notch-1 expression was inhibited by small interference RNA (siRNA), the change of Notch-1, Bcl-2 and Bcl-xL pro- tein level was measured by Western blot. The growth rate of BxPC3 cells was measured by MTT. The change of BxPC3 cells apoptosis was assessed by cell death detection ELISA Kit. Result After inhibiting Notch-1 ex- pression, the growth of BxPC3 cells was inhibited (P 〈 0.01), the level of Bcl-2 and Bcl-xL protein declined at the same time. However, the apoptosis of BxPC3 cells was increased (P 〈 0.01). Conclusion In BxPC3 pan- creatic cancer cells, Notch-1 signaling pathway promotes cell growth and inhibits apoptosis.
