摘 要:目的系膜细胞增殖是多种肾疾病的共同表现,血红素加氧酶-1(heme oxygenase-1,HO-1)可减轻多种疾病的肾损害,文中拟观察不同状态下大鼠肾小球系膜细胞(glomerular mesangial cell,GMC)HO-1的表达,探讨其在系膜细胞增殖中的作用。方法大鼠系膜细胞分为对照组、蟾蜍灵刺激组、血小板衍化生长因子-BB(platelet-derived growth factor-BB,PDGF-BB)刺激组、蟾蜍灵+PDGF-BB刺激组,应用逆转录聚合酶链反应(RT-PCR)技术检测各组细胞HO-1 mRNA的表达,采用Western blot技术检测细胞HO-1蛋白的表达。结果与对照组相比,PDGF-BB刺激组细胞HO-1 mRNA的表达明显减少(P<0.05),蟾蜍灵干预后可抑制PDGF-BB引起的系膜细胞HO-1 mRNA的表达下调(P<0.05),而蟾蜍灵对正常系膜细胞HO-1 mRNA的表达无影响(P<0.05)。Western blot结果显示各组细胞HO-1蛋白的表达与HO-1 mRNA的表达变化趋势一致。结论 PDGF-BB可能通过诱导大鼠系膜细胞HO-1的表达减少引起系膜细胞增殖,蟾蜍灵可抑制此作用,对系膜细胞损伤起保护作用。Objective Glomerular mesangial cell (GMC) proliferation is the common performance of a variety of kidney disea- ses, and heme oxygenase-1 (HO-1) can reduce the renal damage in many diseases. This study aimed to evaluate the role of HO-1 in GMC proliferation. Methods We collected mesangial cells from rats and divided them into a control group, a bufalin group (treated with bufalin at 0.08 p.mol/L), a PDGF-BB group ( treated with platelet-derived growth faetor-BB at 20 ng/ml, and a PDGF-BB + bufalin group (treated with PDGF-BB at 20 ng/ml plus bufalin at O. 08 μmol/L). We detected the expressions of HO-1 mRNA and HO-1 protein in different groups using RT-PCR and Western blot, respectively. Results PDGF-BB significantly reduced the expression of HO-1 at both gene and protein levels ( P 〈 O. 05 ) ; bufalin markedly reversed the reduction ( P 〈 0.05 ), but produced no significant changes in the normal mesangial cells (P 〈 O. 05 ). Conclusion Bufaiin has a protective effect against PDGF-BB-indueed damage to GMCs by up-reg- ulating the expression of HO-1.
