摘 要:目的对微小隐孢子虫(Cryptosporidium parvum,Cp)含EF-hand结构域家族蛋白进行生物信息学研究,探讨该家族蛋白的EF-hand结构域与Ca2+信号的关系以预测该家族蛋白的功能,为隐孢子虫病的诊断和治疗提供新的方向。方法首先通过数据收集,获得EF-hand结构域家族蛋白的序列信息,接着对每个蛋白进行CDD结构域搜索和NCBI BLAST的比对分析,确证为EF-hand结构域家族蛋白,最后进行结构域分析和功能预测。结果搜索到了C.parvumIowa II中18个含有EF-hand结构域的蛋白,它们之间的氨基酸长度,分子量大小以及等电点等都有很大的区别,通过结构域分析把EF-hand家族分成了含一个EF-hand结构域、二个EF-hand结构域、同时含EF-hand和STKc-AGC两种结构域和同时含EF-hand和PKC两种结构域四大类。结论 EF-hand结构域蛋白家族与重要的钙离子信号作用有着密切的关系,而C.parvum对宿主细胞的侵袭能力与胞内的Ca2+水平密切相关,所以EF-hand蛋白家族很有可能成为隐孢子虫病治疗的一个新的突破点。The propose of this study is to conduct bioinformatics research about the EF-hand domain contained protein family of Cryptosporidium parvum , and then predict the relationship between the EF hand domains and the Ca2+ signal which may provide a new direction for the diagnosis and treatment of the cryptosporidiosis. Firstly, based on the data collection, we acquired EF-hand domain contained protein family's sequences. Then the CDD (Conserved Domain Database) domain search and alignment analysis were conducted through the NCBI for every protein to ensure that all of them were EF hand domain con tained proteins. Lastly, we processed the domain analysis and functional prediction. Accordingly, we obtained 18 EF hand do main contained proteins of C. parvum Iowa II, and found that there were significant differences among their amino acid se quence length, molecular weight, isoelectric point, etc. From domain analysis, these 18 proteins could be classified into 4 clas- ses, which were 1 EF hand domain contained proteins, 2 EF-hand domains contained proteins, EF hand and STKc-AGC do- mains contained proteins, and EF-hand and PKC domains contained proteins, respectively. Then we found that this type of protein was closely related to the calcium signals, and the C. parvum's host cell invasion ability had an important relationship with the intracellular Ca4 levels. Hence, the EF-hand proteins family might become a new breakthrough for the treatment of cryptosporidiosis.
