摘 要:目的:研究人羊膜上皮细胞(hAECs)体外诱导分化为唾液腺腺泡细胞样细胞的可能性。方法:分离hAECs并体外培养、鉴定。与8 d龄SD大鼠下颌下腺细胞(SMGCs)在双室共培养系统中共培养。免疫细胞化学、实时荧光定量RT-PCR等方法检测α-淀粉酶及其mRNA的表达。结果:双室共培养1、2周,hAECs中α-淀粉酶mRNA的表达量分别为共培养前的3.38倍及6.6倍。结论:hAECs具有向唾液腺腺泡细胞样细胞分化的能力。Objective: To investigate the differentiation potential of human amniotic epithelial cells(hAECs) into acinar cells in vitro.Methods: hAECs were cultured and identified in vitro.hAECs were cultured on the bottom surface of lower compartment with submandibular gland acinar cells(SMGCs) of 8-day-old SD rat on the microporous membrane in upper compartment using a double chamber system.The expression of α-amylase was determined by immunocytochemical method and fluorescent real-time PCR after 1 and 2 weeks culture respectively.Results: The expression of α-amylase in hAECs was increased by 3.38-fold and 6.6-fold after co-culture for 1 and 2 weeks respectively.Conclusion: hAECs possess the potential to transdifferentiation into acinar cells in vitro.
