辽东楤木叶总皂苷对人胃癌BGC细胞凋亡的影响

摘　　要：目的:探讨辽东楤木叶总皂苷(ETS)对人胃癌BGC细胞凋亡的影响。方法:采用MTT比色法测定辽东楤木叶总皂苷在体外对人胃癌BGC细胞增殖的影响;激光共聚焦显微镜观察FITC-Annexin V/PI双染法检测辽东楤木叶总皂苷作用于人胃癌BGC细胞后引起细胞凋亡的形态特征。结果:MTT比色法结果表明,辽东楤木叶总皂苷各剂量组能不同程度抑制人胃癌BGC细胞的增殖,激光共聚焦FITC-Annexin V/PI双染法检测辽东楤木叶总皂苷可诱导人胃癌BGC细胞出现凋亡早、中、晚期的细胞形态。凋亡早期仅细胞膜呈绿色,细胞核不染色;凋亡中、晚期则细胞膜呈绿色,细胞核呈红色。结论:辽东楤木叶总皂苷能够有效抑制癌细胞增殖、诱导凋亡,具有明显的抗肿瘤作用。Objective：To explore the effect of Aralia Elate Seem Leaf Total Saponin（ETS） on apoptosis of BGC in human gastric cancer.Methods：MTT assay was used to investigate the effect of Aralia Elate Seem Leaf Total Saponin on proliferation in human gastric cancer BGC in vitro.Laser confocal microscope FITC-Annexin V/PI double staining method was used to investigate morphological features of apoptosis caused by Aralia Elate Seem Leaf Total Saponin effect on human gastric cancer BGC.Results：MTT assay results showed that each dose group of the Aralia Elate Seem Leaf Total Saponin could inhibit proliferation in human gastric cancer BGC with different degrees,laser confocal microscope FITC-Annexin V/PI double staining detect the Aralia Elate Seem Leaf Total Saponin could induce apoptosis morphology of human gastric cancer BGC in early,middle and late stage.In early term of apoptosis,cell membrane only was green,nuclear not staining;cell membrane was green and red nucleus in the middle and late stages of apoptosis.Conclusion：The Aralia Elate Seem Leaf Total Saponin can inhibit cancer cell proliferation effectively and induce apoptosis and has an anti-tumor effect significantly.
