摘 要:目的:观察中药抗痫灵对戊四氮点燃癫痫大鼠模型的影响,进一步探讨从调节一氧化氮合酶(nNOS)的nNOS(神经元型)亚型代谢的角度探讨抗痫灵的抗癫痫作用机制。方法:动物模型采用戊四氮点燃大鼠模型。免疫印迹(Western blot)方法观察实验各组大鼠大脑海马nNOS的活性变化。结果:抗痫灵组、丙戊酸钠组、模型组阳性细胞表达与正常对照组相比,有显著性差异(P<0.05),模型组较正常对照组大鼠脑nNOS内活性显著升高(P<0.01),中药组显著低于模型组(P<0.01)。结论:抗痫灵对戊四氮点燃大鼠癫痫发作有显著对抗作用,其作用机制可能与其降低模型大鼠脑内nNOS活性有关。Objective:To observe the effect of Kangxianling on the expression of nNOS in hippocampus of the PTZ - in- duced epilepsy model of rats, we probed into the efficacy and the mechanism of Kangxianling. Methods : PTZ was used to evoke chronic epilepsy model. Ethology changes and electroencephalography were observed after epileptic seizure, expres- sions of nNOS were investigated with Western blot method. Results:The groups of Kanxianling, VPA groups and PTZ- groups were significantly different from the control group ( P 〈 0. 05 ). The groups of Kanxianling was siginificantly differ- ent from the model group( P 〈0.01 ). Conclusion : Kangxianling has anti -epileptic seizure resistant function on PTZ - induced epilepsymodel of rats. Its function mechanism may be to react by reducing the content of nNOS in the hippocampus.
