摘 要:对山羊痘重组沙门菌S7207/pVAX1-P32疫苗株的安全性、稳定性及S7207/pVAX1-P32在动物体内散布规律进行测定。选择60只小鼠,随机分为4组,1组为生理盐水对照组,2组~4组为试验组,用上述沙门菌株S7207以108、109、1010cfu的剂量口服免疫小鼠,2周后相同剂量加强免疫,通过在不同时间段称量小鼠体重显示,试验组小鼠生长情况与对照组无明显差异,不同组别小鼠大体及内脏器官也未发生病变,说明S7207/pVAX1-P32安全性良好;通过PCR方法检测P32基因在体内各组织分布情况,在7d后体内各组织中都检测出目的基因,说明DNA在小鼠体内广泛分布。体外传代含pVAX1-P32和不含pVAX1-P32菌株,含质粒菌株经60代传代后含重组质粒菌的比例在50%以上,传代至120代时仍在10%以上,而不含质粒菌株明显低于含质粒菌株,结果证明含质粒菌株稳定性较好。研究结果提示,利用减毒沙门菌为载体传递S7207/pVAX1-P32疫苗具有良好的安全性和稳定性,其质粒在小鼠体内分布广泛,对未来口服疫苗的使用提供了参考依据。To observe the safety and stability of the oral vaccine prepared with recombinant goat-pox Salmo- nella S7207/pVAX1-p32 and to explore the distribution in vivo, 60 mice were divided randomly into 4 groups. Group 1 was injected with PBS as the control. Group 2-4 as experimental groups were orally inoc- ulated with a dose of 10^8、10^9、10^10cfu per mouse. Booster immunization was performed with the equal dose 2 weeks later. The results showed that no significant difference was observed between the control and the experimental groups in growth, and no lesions were found. Furthermore, P32 gene was detected from ew ery tissue by PCR 7 days after the experiment. Moreover, the bacterial strain was continuous cultured in- cluding the plasmid pVAXl-p32 involved and uninvolved. The recombinant plasmid accounted for more than 50 percent of all the experimental strain at 60 generation, with more than 10% at 120 generation, which showed a significantly higher ratio than the plasmid-exclusive strain. From all the evidence, we could build up the indication that the S7202/pVAX1-p32 vaccine had a wide distribution in vivo, with the reliable safety and stability, which confer a base reference to the employment of the oral vaccine.
