七味白头翁汤对小鼠结肠炎作用的研究

摘　　要：[目的]通过动物实验观察七味白头翁汤灌胃治疗溃疡性结肠炎的疗效,并探讨其可能的作用机理。[方法]130只体重18～20g的雄性清洁级BALB/C小鼠随机分为模型组(30只)、中药组(30只)、西药组(30只)、乙醇对照组(20只)、正常对照组(20只)。除正常对照组和乙醇对照组外,其余三组采用经典的噁唑酮+无水乙醇法制备溃疡性结肠炎模型。造模成功后,正常对照组、乙醇对照组及模型组每天予0.9%氯化钠灌胃,而西药组和中药组分别予以柳氮黄氨吡啶和七味白头翁汤悬浊液灌胃治疗,灌胃1周。每天记录各组小鼠的体重、大便性状及便血情况,粪隐血检测采用联苯胺法,并采用疾病活动指数(DAI)对动物状态进行评分,1周后处死所有小鼠,观察其结肠组织病理学变化及结肠黏膜IL-5和IL-13 mRNA表达情况。[结果]中药七味白头翁汤能显著改善溃结小鼠体重及活动状态,修复结肠组织的病理损害,有效降低IL-5和IL-13的表达水平。[结论]七味白头翁汤对溃疡性结肠炎急性期有良好疗效,能够减轻肠道炎症,修复黏膜损伤和促进溃疡愈合。[ Objective] To investigate the inhibitory effects and mechanisms of Qiweibaitouweng decoction on ulcerative colitis in mice. [ Method] 130 Clean BALB / C mice were randomized into five groups： model group, Chinese medicine treatment group, Western medicine treatment group , normal group and Ethanol con- trol group. Except the mice in normal group and Ethanol control group, all mice were induced by oxazolone enema（ dissolved in 100% ethanol）. The western medicine treatment group was given Sulfasalazine （SASP） by stomach lavage, Chinese medicine treatment group was given Qiweibaitouweng decoction by stomach lay- age, The remaining three groups was given normal saline by stomach lavage, for one week. A daily record of mice weight, stool and hemafecia condition, fecal occult blood detection by benzidine method, and evaluating the disease activity index （ DAI ） to animal status. One weeks later, all the mice were killed and colon tissue preparation. To observe the histopathological changes of the colon, and determine the expression and compare the changes of IL - 5 mRNA and IL - 13 mRNA of colon in mice. [ Result ] Qiweibaitouweng can significantly improve mice weight and activity state, repair colonic tissue pathological damage, and reduce the expression levels of IL-Sand IL- 13 effectively. [ Conclusion] Qiweibaitouweng decoction exerts protective effect in ex- perimental colitis, the effect seems to be related to relieving inflammatory reaction, repairing mucosal injury and healing ulcer.
