摘 要:[目的]观察补肾通窍方对糖尿病大鼠丘脑前背侧核神经元Bax与Bcl-2表达的影响。[方法]采用腹腔内注射链脲佐菌素55mg/kg制作糖尿病大鼠模型,成模4周后灌注固定,取脑作连续石蜡冠状切片,用免疫组化SABC染色,观察ad神经元Bax与Bcl-2的表达。[结果]糖尿病模型组较正常对照组Bax阳性细胞密度增多(P<0.01),Bcl-2阳性细胞密度减少(P<0.01)。补肾通窍方治疗组和胰岛素治疗组Bax阳性细胞密度较糖尿病模型组减少(P<0.01),而Bcl-2阳性细胞密度增多(P<0.01)。补肾通窍方治疗组和胰岛素治疗组的Bax、Bcl-2阳性细胞密度无显著差异(P>0.05)。[结论]补肾通窍方能抑制糖尿病大鼠ad神经元Bax的表达,维持Bcl-2的正常表达;补肾通窍方可以防治糖尿病大鼠ad神经元的退行性变。[Objective]To investigate the effect of bushen tongqiao decoction(BSTQD) on Bax and Bcl-2 expression of anterodorsalthalamic nucleus (Ad) in diabetes rats. [Methods] Diabetes was induced with streptozotocin in rats. Four weeks later, rats were transcardially perfused and routine paraffin was made in coronal sections in brains, and then stainning of SABC was used to observe the expression of Bax and Bcl- 2 in Ad. [Results] The density of the Bax-positive cells was increasing significantly, while the Bcl-2-positive cells decreasing. The density of bax- positive cells in rats of both BSTQD group and insulin group were significantly lower than the model group, while the Bcl-2-positive cells higher. No significant difference was found between BSTQD group and insulin group. [Conclusion] BSTQD can restrain the expression of Bax of Ad in diabetes rats and maintain the expression of Bcl-2. BSTQD can prevent recession of Ad in diabetes rats.
