摘  要:目的探讨两种实验性糖尿病大鼠不同频次低糖血症时血胰高血糖素的变化特点及差异。方法不同饲养方式结合不同剂量链尿佐菌素腹腔注射诱导制备1型和2型糖尿病大鼠模型。正常、1型和2型糖尿病大鼠随机分为对照组(NC、T1C、T2C),一次低糖血症组(NL1、T1L1、T2L1)和二次低糖血症组(NL2、T1L2、T2L2)。低糖血症组大鼠用胰岛素诱导低糖血症。所有大鼠在腹腔注射9g/L氯化钠注射液或注射胰岛素前、注射后1.5h及注射后4.5h心脏采血测定血糖值后离心取血清,采用ELISA试剂盒测定胰高血糖素。采用重复测量数据的方差分析进行数据分析。结果 NL1组和NL2组在1.5h胰高血糖素达峰值,T1L1组、T1L2组和T2L2组均在4.5h胰高血糖素达峰值。各组糖尿病大鼠晚期(4.5h)胰高血糖素与血糖比值均较早期(1.5h)显著升高(F=13.746~29.377,q=5.854~13.681,P<0.05、0.01);T1L1组和T1L2组1.5h胰高血糖素与血糖比值与正常大鼠比较均显著降低(F=14.572,q=6.233~7.109,P<0.05),4.5h升高但差异无显著性(P>0.05);T2L1组和T2L2组1.5、4.5h均显著降低(F=19.172~24.351,q=9.237~11.377,P<0.01)。与1型糖尿病大鼠比较,2型糖尿病大鼠一次低糖血症发作时仅4.5h胰高血糖素与血糖比值显著降低(F=18.409,q=8.966,P<0.01),二次低糖血症发作时1.5h和4.5h均显著降低(F=14.391~15.003,q=6.244~6.951,P<0.05)。相同类型糖尿病大鼠与一次低糖血症比较,二次低糖血症后T1L2组1.5、4.5h胰高血糖素与血糖比值均明显升高(F=16.479~17.287,q=7.142~8.031,P<0.05),T2L2组仅4.5h明显升高(F=13.977,q=5.833,P<0.05)。结论两种糖尿病大鼠均存在胰高血糖素对低糖血症的反向调节障碍且存在差异,以低糖血症发生后早期阶段为著,2型糖尿病大鼠较1型糖尿病大鼠更严重。Objective To investigate the difference of serum glucagon response to different frequency of hypoglycemia in two kinds of diabetic rats. Methods Type 1 and type 5? diabetic models in rats were created by administrating different feeding and different doses of streptozotocin intraperitoneal injection. Normal rats, TIDM rats and T2DM rats were randomized to control group (NC, T1C, T2C), hypoglycemia group (NL1, T1L1, T2L1) and recurrent hypoglycemia group (NL2, T1L2, T2L2), re- spectively. The occurrence of hypoglycemia was induced by injecting insulin. Blood samples were obtained from heart before perito- neal injection (insulin or 9 g/L sodium chloride solution) and 1.5 h and 4.5 h after the injection. After blood glucose being detected by glucometer, the blood serum was extracted by centrifugation and kept for subsequent glucagon determination with ELISA kit. ANOVA for the repeated measures was used for data analysis. Results The time of reaching the peak concentration of glueagon was as follows: in groups NL1 and NL2, in 1.5 h, while in groups T1L1, TIL2 and T2L2, in 4.5 h. Ratio of glueagon and blood glucose in advanced stage (4.5 h) in each group was higher than that in early stage (1. 5 h) group (F= 13. 746-29. 377;q= 5. 854 13. 681 ;P(0.05,0.01). Compared with normal rats, the ratio decreased in T1L1 and T1L2 group at 1.5 h (F= 14. 572, q=6. 233-7. 109,P〈0.05), increased at 4.5 h, but with no statistical difference (PD0.05) ; an obvious decrease was also found inT2L1 andT2L2 group at 1.5 hand4.5 h (F=19.172 24. 351,q=9.237-11.377,P〈[0.01). Compared with type 1diabetic rats, the ratio of type 2 diabetic rats was found significantly decreased at 4.5 h in the first hypoglycemia (F= 18. 409, q= 8. 966, P〈0.01), it also significantly decreased at both 1.5 h and 4.5 h in recurrent hypoglycemia (F= 14. 391 - 15. 003, q= 6. 244 - 6. 951 ,P〈0.05).
