摘  要:目的研究附睾特异表达的RNase9蛋白在精子获能过程中的作用及对精子运动的影响。方法本实验主要采用免疫荧光技术、金霉素染色法以及计算机辅助精子分析法研宄RNase9蛋白或抗体对精子的作用。结果精子在获能过程中RNase9蛋白的定位不断发生改变,顶体反应发生后,精子头部的RNase9蛋白荧光信号明显减弱或消失。精子与RNase9蛋白共同孵育后,精子运动能力也明显降低。结论RNase9蛋白参与精子获能过程并在体外抑制精子的运动。Objective To investigate the role ofepididymis-specific protein RNase9 during sperm capacitation and its effect on sperm motility. Methods Immunofluorescence technique, CTC staining and computer assisted sperm analysis were used respectively to evaluate the biological function of epididymis-specific protein RNase9 on sperm. Results The location of RNase9 protein in the surface of sperm was continuously changed during capacitation. The fluorescence signal of RNase9 protein in head of sperm decreased or diminished after acrosome reaction. Sperm motil- ity was inhibited under with RNase9 protein. Conclusion RNase9 protein might be involved in sperm capacitation and inhibit sperm motility in vitro.
