摘 要:初步探讨重组生长激素 (rGH )改善肝切除术后疲劳综合征 (POF )的分子机制及对肝脏再生的影响。方法 采用半定量逆转录聚合酶链式反应 (RT PCR )技术 ,分别对治疗组及对照组大鼠切肝 4 0 %术后 3 ,7d生长激素受体mRNA (GHRmRNA )的表达及相对肝重量 (肝重 体重 )进行研究。结果 手术后 3d组GHRmRNA表达量明显下降 ;治疗组术后3dGHRmRNA的表达和术后 7d肝脏再生较对照组明显增加 (P <0 .0 5)。结论 术后GHRmRNA的低水平表达 ,可能是导致POF发生的机制之一 ;Objective To investigate the molecular mechanism of recombinant growth hormone(rGH) in improving the postoperative fatigue syndrome(POF) and affecting liver regeneration after hepatectomy. Methods Expression of growth hormone receptor mRNA(GHR mRNA) was examinated with reverse transcription polymerase chain reaction(RT PCR) in rGH treatment group(rGH group) and control group after partial hepatectomy. Results (1) Significantly low expression of GHR mRNA could be observed 3 days after operation in the 2 groups. (2) In rGH group, GHR mRNA 3 days after operation and liver regeneration 7 days after operation were significantly increased( P <0.05) compared with control group. Conclusions Postoperative low expression of GHR mRNA may be one of the reasons which lead to POF; rGH can enhance postoperative expression of liver GHR mR NA and liver regeneration.
