大鼠主动全身过敏试验方法的探讨

摘　　要：目的建立大鼠主动全身过敏试验模型,评价中药注射剂过敏反应。方法选用豚鼠和BN、F344、SD、Wistar4种品系大鼠,以卵白蛋白(OVA)、天花粉蛋白(TCS)、清开灵注射液、注射用灯盏花素为受试物,分别隔日腹腔注射致敏,共3次,于末次致敏后第14天静脉注射2倍致敏剂量激发。同时设生理盐水对照组。观察各组动物的过敏反应发生率及过敏反应症状。结果阳性药卵白蛋白和天花粉蛋白可引起豚鼠和大鼠发生过敏反应,发生率均为100%,豚鼠发生的过敏反应症状程度略高于大鼠。清开灵注射液可引起豚鼠和BN大鼠发生过敏反应,发生率分别为50%和100%。注射用灯盏花素导致F344、BN和SD大鼠发生过敏反应,发生率分别为100%、100%、83.33%,且F344大鼠呈现强阳性反应,而豚鼠则呈现阴性反应。结论大鼠可推荐作为主动全身过敏试验的动物模型之一,对致敏原的敏感性在动物不同品系间可能存在差异。AIM To establish an active systemic anaphylaxis（ASA） test rat model for assessing allergic response induced by traditional Chinese medicine injections.METHODS ASA was performed by using guinea pigs,BN,F344,SD and Wistar rats,with ovalbumin,trichosanthin,Qingkailing Injection and Breviscapine Injection as test substances.The experimental animals were intraperitoneally injected with test substances respectively every other day for 3 times.On the fourteenth day,after the last senistization,the experimental animals were injected with the same medicine in the dosage of twice as much as sensitization dose through vein,while the control group was injected with sterile saline at the same volume.The symptoms and the incidence of allergic response were assessed according to ASA symptom scale and evaluation criteria.RESULTS The guinea pigs and the rats of four strains showed positive allergic response to ovalbumin and trichosanthin with 100% incidence.The degree of allergic response symptoms of guinea pigs was slightly higher than that of rats.Qingkailing Injection induced allergic responses in guinea pigs and BN rats with 50% and 100% incidences,respectively.Breviscapine Injection caused allergic responses in F344,BN and SD rats with 100%,100% and 83.33% incidences,respectively,while F344 rats presented strongly positive response.Guinea pig was negative reaction on Breviscapine Injection.CONCLUSION Rats can be used to establish a suitable animal model in ASA test.The difference between rat strains may exist in the allergic response to the allergen.
