高分离度快速液相色谱法测定小儿清热利肺口服液中绿原酸和牛蒡苷的含量

摘　　要：目的:建立同时测定小儿清热利肺口服液中绿原酸和牛蒡苷含量的方法。方法:采用高分离度快速液相色谱法。色谱柱为ZorbaxSB-C18柱,流动相为0.1%冰醋酸溶液-乙腈(梯度洗脱),检测波长0～5.5min为327nm,5.5～7.0min为280nm,流速为1.0mL.min-1,柱温为20℃,进样量为1.0μL,采集频率为1s。结果:绿原酸、牛蒡苷进样量分别在0.014602～0.29204、0.051396～1.02792μg范围内与峰面积积分值有良好的线性关系(均r=0.9999);平均加样回收率分别为99.56%、99.96%,RSD分别为1.22%、1.09%(n=6)。结论:本方法操作简便易行,可在保持分离度的前提下缩短分离时间,减少溶剂消耗,节约分析成本,提高日常检验工作的效率,可用于控制该药的生产工艺及质量。OBJECTIVE:To establish the method for the content determination of chlorogenic acid and arctiin in Xiaoér qingre lifei oral solution.METHODS:Rapid resolution liquid chromatography(RRLC) method was adopted.The separation was performed on Zorbax SB-C18 column with mobile phase consisted of 0.1% acetic acid-acetonitrile(gradient elution) at the flow rate of 1.0 mL·min-1.The detection wavelength was set at 327 nm during 0～5.5 min and 280 nm during 5.5～7.0 min.The column temperature was kept at 20 ℃ and injection volume was 1.0 μL.Response time was 1 s.RESULTS:The linear ranges of chlorogenic acid and arctiin were 0.014 602～0.292 04 μg and 0.051 396～1.027 92 μg,respectively(r=0.999 9).The average recoveries were 99.56%(RSD=1.22%,n=6) and 99.96%(RSD=1.09%,n=6),respectively.CONCLUSION:The developed method is simple,feasible.It shortens the separation time on the premise of the maintenance of separation rate,decreases solvent consumption and reduces the cost without reducing the efficiency,it can be used for quality control in Xiaoér qingre lifei mixture.
