益气软坚散结法对甲减大鼠心肌TRα1mRNA表达的影响

摘　　要：目的:研究益气软坚散结法对甲状腺功能减退大鼠心肌TRα1mRNA表达的影响。方法:(1)将Wist-ar大鼠随机分为正常组、模型组、L-T4组、益气软坚散结组(自拟芪贝复元饮)4组,每组30只。(2)正常组:普通饲料,双蒸水;模型组:低碘饲料,双蒸水。(3)酶联免疫吸附测定法(Elisa法)测大鼠血清促甲状腺激素(TSH),放射免疫分析法(RIA)测血清总T3(TT3),总T4(TT4)。实时荧光定量PCR(Real-time PCR法)检测大鼠心肌TRα1mRNA的表达。砷铈分光光度法测定尿碘含量。光镜下观察大鼠心肌心肌形态。心电图机记录大鼠心电变化。结果:56天时予处理因素的L-T4组和芪贝复元饮组大鼠血清TT3值均升高(P<0.01),二组之间比较无差异;血清TT4值均升高(P<0.001),二组之间比较无差异;TSH值明显降低(P<0.01),其中较L-T4组血清TSH值降低明显。心电图描记发现芪贝复元饮组与L-T4组比较有差异(P<0.05),心率及低电压表现有恢复;芪贝复元饮组心肌TRα1mRNA的表达较L-T4组高0.81倍(P<0.05);心肌细胞形态观察发现芪贝复元饮组较L-T4组恢复的明显。结论:与L-T4比较益气软坚散结法能使甲减心肌损害得到恢复,其作用机制与TRα1mRNA在心肌细胞上的表达有关。Objective：To study the effect of Yiqi Ruanjian Sanjie method on myocardial TRα1mRNA expression of Wistar rats with hypothyroidism.Methods：Wistar rats were divided into 4 groups：normal group,model group,L-T4 group,and Yiqi Ruanjian Sanjie method.Each group had 30 rats.Normal group：ordinary diet and double distilled water（DDW）were fed while model control group was fed with low iodine diet（LD）and DDW.Serum thyroid-stimulating hormone（TSH）was measured with Elisa.Serum total levels of T4（TT4）and T3（TT3）were measured with radioimmunoassay（RIA）.Myocardial TRα1mRNA expression was measured with real-time quantity PCR.Ger-Arsenic cerium spectrophotometry was used to measure median urinary iodine（MUI）.Myocardial morphological changes were observed under optical microscope,and myocardial electrocardio was measured by ECG machine.Results：Giving the processing factor 56 days later,serum TT3 values in L-T4 group and Yiqi Ruanjian Sanjie group were higher than that in normal group and model group（P 〈0.01）.Serum TT4 value was higher（P〈 0.001）.Serum TSH value was lower（P 〈0.01,and Yiqi Ruanjian Sanjie group was obviously lower than L-T4 group）.ECG indicated differnece between L-T4 group and Yiqi Ruanjian Sanjie group（P 〈0.05）,and heart rate and low-volume were recovered.Yiqi Ruanjian Sanjie group of myocardial TRα1mRNA expression was 1.08 times higher than L-T4 group.Myocardial morphology of Yiqi Ruanjian Sanjie group was recovered obviously more than L-T4 group.Conclusion：Compared with L-T4 group,Yiqi Ruanjian Sanjie method can make myocardic of hypothyroidism recovery by myocardial TRα1mRNA.
