威柏膏外敷治疗大鼠急性痛风性关节炎的实验研究

摘　　要：目的:探讨威柏膏外敷治疗急性痛风性关节炎模型大鼠的药效及可能的作用机制。方法:雄性Wist-ar大鼠随机分为正常对照组(A)、模型组(B)、秋水仙碱组(C)、威柏膏外敷组(D)、秋水仙碱加威柏膏组(E)。观察大鼠足功能变化,检测其受试关节周径、滑膜病理改变,以及关节滑膜组织中白介素1-β(IL-1β)、5-羟色胺(5-HT)和TLR-4(Toll-like receptor 4)的相对表达量。结果:与模型组比较,各治疗组均可减少炎性细胞浸润,并能显著降低IL-1β、TLR-4的表达。E组较D组降低IL-1β的表达更为显著;E组能显著降低5-HT的相对表达含量。结论:威柏膏外敷能下调大鼠急性痛风性关节炎的炎性反应,止痛消炎,其机制可能是降低了滑膜组织中IL-1β、TLR-4及5-HT的表达;威柏膏联合秋水仙碱比单用两者之一的效果更好。Objective： To investigate the effects of WeiBai Balm on acute gout in synovial tissue in Wistar rats.Methods： 40 male Wistar rats were randomly divided into normal control group（A）,acute gout model group（B）,colchicine group（C）,WeiBai Balm group（D）,colchicine combined with WeiBai Balm group（E）.Acute gout model was constructed according to Coderre classical method.Gait and joint perimeter were observed,and the expressions of interleukin-1β,serotonin,and Toll-like receptor-4 in synovial tissue of joint were measured by immunohistochemistry.Results： Compared with normal control,the synovial tissue of model rats were obviously hyperplastic,a great deal of neutrophils and macrophagus infiltrated and blood capillaries distended and congested.Compared with model rats,the infiltration of inflammatory cells was decreased and the expressions of interleukin-1β and Toll-like receptor-4 were significantly down-regulated in C,D,E groups（P〈0.01）.The expression of serotonin was reduced dramatically in E group（P〈0.05）.C,D,E groups functioned almost equally in the expression of Toll-like receptor-4.Conclusion： WeiBai Balm could attenuate inflammation and alleviate pain of acute gout,possibly through down-regulation of the expressions of interleukin-1β,serotonin,and Toll-like receptor-4 in synovial tissue of joint.WeiBai Balm combined with colchicine was more effective than single WeiBai Balm or colchicines treatment.
