阿月浑子杂种幼胚培养技术的研究

摘　　要：以阿月浑子杂种果实和人工授粉后120d的杂种幼胚为试材,对杂种果实生长发育规律及幼胚培养进行了研究,探讨了冷藏处理时间、不同光照条件和培养时间对幼胚萌发和成苗的影响。结果表明:阿月浑子人工授粉80d后,单果鲜重和PF值仅为1.27g和0.24;授粉后100d,单果鲜重和PF值为1.57g和0.43;授粉后120d,单果鲜重和PF值为2.81g和0.80;3种冷藏处理中,以冷藏处理90d时幼胚萌发及生长效果最好,幼胚萌率为91.67%,平均真叶数为6.09个,平均苗高为4.14cm;在培养方式上,以黑暗处理后培养20d效果最好,幼胚萌发率为86.67%,平均真叶数为5.38,平均苗高为4.23cm。In this experiment, hybrid fruits and immature embryos of Pistacia vera L., growing for 120 d after artificial pollination, were used as materials. The growth rule and immature embryo culture of pistachio＇s hybrid fruit were studied. The effects of cold treat-ment time, different light conditions and culture time on immature embryo development and normal seedling rate were also investigated, indicating that the single--fruit fresh weight and PF value were only 1.27 g and 0.24 when growing for 80 d after artificial pollination, reaching 1.57 g and 0. 43 in 100 d after, with the average fruit fresh weight and PF value being 2.81 g and 0.80 respectively, in 120 d after. The suitable cold treatment time for immature embryo ger-mination and growth is 90 d,the rate of embryo germination is 91. 67%, the average leaf number is 6.09 and the average seedling height is 4. 14 cm. It was suggested that the best culture way is the dark treatment for 20 d, the rate of embryo germination is 86.67%, the average leaf number is 5.38 and the average seedling height is 4.23 cm.
