摘 要:目的探讨不同浓度的红景天苷对大鼠肾脏缺血再灌注损伤的保护作用以及HSP72表达水平的关系。方法将48只健康sD大鼠随机分为6组,分别为红景天苷5mg/kg组、10mg/kg组、20mg/kg组、40mg/kg组、假手术组和空白对照组。通过切除有肾,夹闭左肾蒂45min,建立肾脏缺血再灌注模型。分别检测大鼠血清肌酐(Scr)、尿素氮(Bun)、肾组织常规病理及肾组织中HSP72的表达。结果与红景天苷5mg/kg组、10mg/kg组、40mg/kg组相比,红景天苷20mg/kg组Scr水平、Bun水平明显下降(P〈0.01),病理损伤明显减轻,组织中HSP72表达也明显减少。与空白对照组比较,红景天苷20mg/kg组Scr水平、Bun水平略有上升,病理损伤较轻。结论应用红景天苷后,大鼠肾脏缺血再灌注损伤明屁减少,其机制与增强肾脏抗氧化能力,减轻肾脏组织炎性细胞浸润和抑制肾组织HSP72表达有关,其中20mg/kg浓度的红景天苷保护效果最佳。Objective To study the effect of salidroside in rat renal ischemia reperfusion injury and the relationship between the expression levels of HSP72. Methods The 48 healthy SD rats were randomly divided into six groups, namely salidroside 5mg/kg group, 10mg/kg group, 20mg/kg group, 40mg/kg group, operation group and control group. Through the removal of the right kidney, left renal pedicle clamping for 45 minutes to establish renalischemia-reperfusion model. Were measured in serum creatinine (Scr), blood urea nitrogen (Bun), normal kidney and renal pathology in the expression of HSP72. Result Comparison group of salidroside 5mg/kg, 10mg/kg group, 40mg/kg group, the level of salidroside 20mg/kg group Scr, Bun levels were significantly decreased (P〈0.01), significantly reduced pathological damage, tissue expression of HSP72 also decreased significantly. Compared with the control group, salidroside 20 mg/kg group Scr levels, Bun level increased slightly, less pathological damage. Conclusion Application of salidroside after renal ischemia-reperfusion injury significantly reduces, the mechanism and enhances the antioxidant capacity of the kidneys, reducing kidney tissue inflammatory cell infiltration and inhibition of HSP72 expression in renal tissue,with concentrations of 20mg/kg salidroside the best for protection.
