摘 要:[目的]研究秦皮乙素的最佳提取分离工艺。[方法]利用HPLC法测定秦皮中秦皮乙素的含量,以秦皮乙素含量为指标,用L9(34)正交试验法来确定最佳提取工艺;以秦皮乙素的分离情况为指标,采用聚酰胺柱筛选出最佳纯化分离工艺。[结果]最佳提取工艺为:料液比1∶8(W/V,g/ml,下同),乙醇浓度95%,提取3次,每次1.5 h。聚酰胺柱对秦皮乙素的分离效果较好,纯化的最佳工艺为:吸附流速为8 BV/h,上样液与树脂的体积比约为8∶1,以蒸馏水洗脱,洗脱流速为4 BV/h,收集洗脱液;在此条件下,秦皮乙素的总洗脱率达到90%以上,精制率均达到50%以上。[结论]试验所选择的秦皮乙素的提取分离方法效果较好。[Objective] To study the optimal extraction and purification process of esculetin from CORTEX FRAXINI.[Method] Using the content of esculetin measured by HPLC as the index,the optimal process of extracting esculetin was determined by L9(34) orthogonal test.With the isolation of esculetin as the index,the optimum isolation and purification process was determined by polyamide column.[Result] The optimal extraction process was extracting three times,each extraction 1.5 h,with eight-fold amount of 95% ethanol.Polyamide column could effectively isolate esculetin by following the steps below: the test solution was loaded on polyamide column at an adsorption flow rate of 8 BV/h and eluted with distilled water at an elution flow rate of 4 BV/H.The volume ratio of loading solution to resin was set at 8∶ 1 g/ml(W/V).Under the above conditions,the total elution rate and refining rate of esculetin were more than 90% and 50%,respectively.[Conclusion] The method used in the study is good for extracting esculetin from CORTEX FRAXINI.
