摘  要:目的观察C型利钠肽(CNP)在单侧输尿管梗阻(UUO)模型大鼠肾脏的表达特点和对细胞外基质(ECM)代谢的调控作用。方法健康雄性8周龄Wistar大鼠24只,随机分为UUO组和假手术组并建立模型,于术后1 d和3个月处死大鼠,检测尿蛋白浓度和血清学指标,HE染色观察肾组织形态学变化,免疫组化法、荧光实时定量PCR法检测肾组织CNP、胶原IV(Col-IV)、基质金属蛋白酶(MMP-2、MMP-9)及其组织抑制因子(TIMP-1)的表达情况。结果术后3个月,与假手术组比较,UUO组大鼠血浆白蛋白降低,肌酐、尿素氮、尿蛋白均升高(P<0.05);CNP、MMP-9、MMP-2蛋白及mRNA表达均下调(P<0.05);Col-IV、TIMP-1蛋白及mRNA表达均上调(P<0.05)。结论 CNP表达减少可能是引发ECM代谢失衡,最终导致肾小管间质纤维化的重要原因。Objective To explore the expressive characterization of C-type natriuretic peptide(CNP) and its modulation of extracellularmatrix(ECM) metabolism in the kidney of unilateral ureteral obstruction(UUO) model rats.Methods Twenty four healthy male Wistar rats were randomly divided into two groups: UUO group and sham group.UUO model was established and all rats were killed at the 1st day and the 3th month.In both groups,the proteinuria and plasma levels were examinated.Renal morphological changes were observed by HE staining.Expression of CNP,MMP-2,MMP-9,TIMP-1,Col-IV were observed by immunohistochemistry,real time-PCR.Results At the 3th month,compare with the sham group,proteinuria levels,albumin,serum creatinine,blood urea nitrogen levels increased in the UUO group(P 〈 0.05).Immunohistochemical assay and real time-PCR revealed that the expression of CNP,MMP-2,MMP-9 decreased and TIMP-1,Col-IV increased in the renal tissue of UUO rats(P 〈 0.05).Conclusion The CNP expression decreases in the kidney of UUO rats,which may be caused the imbalance of ECM and tububinterstitial fibrosis.
