摘 要:目的探讨原发性高血压(EH)患者G蛋白β3亚单位基因的C825T多态性与美托洛尔降压疗效的关系。方法 EH患者52例,口服美托洛尔25mg/次,每天2次,连续服用4周。采用基因芯片技术进行基因多态性检测,分析不同基因型之间血压下降值的关系。结果服用美托洛尔治疗4周后,CT+TT基因型患者(43例)收缩压的下降值为(21.07±12.04)mm Hg,平均动脉压的下降值为(13.33±7.31)mm Hg,均明显大于CC基因型患者(9例)的(9.44±14.65)mm Hg和(7.07±7.34)mm Hg(P<0.05)。但两组之间舒张压下降值无统计学差异。结论 G蛋白β3亚单位基因的C825T多态性与美托洛尔降压疗效相关。携带T等位基因的患者收缩压及平均动脉压下降更明显。Objective To investigate the correlation of G protein β3 subunit gene C825T polymorphism and efficacy of metoprolol in treating essential hypertension(EH). Methods Fifty-two elder patients with EH were teated with metoprolol 25 mg,bid for 4 weeks. The G protein β3 subunit gene C825T polymorphism was genotyped by gene chip technique, The decrease of systolic blood pressure(SBP) and mean arterial pressure(MAP) were compared between two genotype groups of CT +TT and CC. Results The reductions in SBP and MAP of CT+TT genotype group were (21.07± 12.04) mmHg and (13.33±7.31) mmHg,respectively,which were significantly more than (9.44± 14.65) mmHg and (7, 07±7.34) mmHg of CC genotype group(P〈0. 05). There was no significant difference in the reduction of diastolic blood pressure between groups of CT±TT and CC genotype. Conclusion The G protein β3 subunit gene C825T genotype is associated to the hypotensive efficacy of metoprolol. The reductions in SBP and MAP are greater in EH patients carrying T allele.
