转染病毒白细胞介素10基因对同种异体大鼠皮肤移植术后皮肤CD80和CD86表达率的影响

摘　　要：目的探讨免疫调节因子vIL-10基因对修复同种异体大鼠全层皮肤缺损的免疫耐受作用，以期为解决皮肤移植领域的免疫排斥问题提供理论依据，为解决大面积皮肤缺损病人皮源不足问题奠定必要的基础。办法将转染vIL-10的骨髓基质干细胞经腹腔注射输入实验组大鼠体内，以病毒白细胞介素10表达阳性的大鼠为受体，同种异体大鼠为供体，对照组为注射未转染vIL-10的骨髓基质干细胞的大鼠，空白组为注射同量生理盐水的大鼠，行同种异体全层皮肤移植。取3组1，2，3周的皮肤标本制成石蜡切片，进行免疫组织化学检测并拍照。观察在vIL-10的作用下CD80，CD86的表达率变化。结果CD80，CD86表达率不断升高，但实验组表达率较同期对照组、空白组明显降低（P〈0．05），同期对照组、空白组表达率无明显差异。结论转染病毒白细胞介素10能够降低CD80，CD86的表达水平进而影响T淋巴细胞分型，从而降低机体的免疫排斥反应。Objective To investigate the immune tolerance induction of inhibitory and imnmne regulatory cytokine vIL-10 on repair of full-thickness skin defect from allograft rats to solve the problem of immune rejection and provide a theoretical basis for solving large skin defects of the patients＇paper source shortage and lay the necessary hmndation. Methods The MSCs expressing vIL-10 were injected into the abdominal of the rats. ＇Fake the rats injected with MSCs expressing vIL-10 as the experimental group,take the rats injected with MSCs not expressing vIL-10 as the control group,and take the rats injected with physiological saline as the sham group. Allogenic full-thickness skin graft were carried out among the three group. The skin samples of three group in 1,2,3 week were made into paraffin sectinn and checked by imnmnohistnchemistry and dyed by PV6001 method. To observe the positive rate change of CD80, CD86 under the influence of vIL-10. Results The positive rate of CD80 and CD86 were increased gradually in the process of rat allogenic skin transplantion rejection. And the positive rate of experimental group was significant lower than control group and sham group in the same period（ P 〈 0.05 ）. The positive rate of control group and sham group showed that there was no difference. Conclusion The interleukin 10 can reduce the level of CDS0/CD86 and effect the sonmtotype of T lymphocytes,and then decrease the organism immunological rejection.
