摘 要:为研究低氮营养对烟草生长的主要影响,分别在移栽后50 d到70 d期间测量了低氮胁迫和正常营养条件下烟株中部叶片的叶宽、叶长和50 d与60 d时的株高,同时应用实时荧光定量PCR方法分析了移栽后50 d到70 d期间烟草叶片中胁迫响应基因亲环素mRNA的表达量。结果发现低氮营养下叶长、叶宽和株高都小于正常营养条件下的叶片,但是叶长、叶宽平均生长增幅显著大于正常营养下的叶片,而株高的增长幅度则没有明显差异。低氮胁迫下叶片亲环素基因mRNA表达量与正常条件下的基因转录表达量相比有显著增加,最高为正常条件下的6.9倍,并且在移栽后50 d到70 d期间相对转录水平呈递增趋势。水培实验也证明低氮胁迫可显著诱导叶片亲环素基因的转录表达,诱导表达量达2.0倍之多。结果表明低氮营养胁迫对烟草叶片的生长影响较为显著,且低氮营养可以诱导烟草叶片亲环素基因mRNA表达大量增加,推测低氮营养胁迫下亲环素基因的大量诱导表达可能参与了对烟草叶片生长的调节过程。The effects of low nitrogen nutrition on tobacco growth was investigated. Characters such as leaf width, leaf length of middle-stalk leaves were measured from 50 days to 70 days after transplanting together with plant heights at 50 days and 60 days. The mRNA expression level of a stress-responsive signal cyclophilin gene was analyzed by real-time quantitative PCR. Results showed that leaf width, leaf length and plant height are smaller in low nitrogen group com-pared with that grown under normal nitrogen. Leaf growth increments under low nitrogen condition were larger than that of normal nitrogen. No significant differences between increments of plant heights under two nitrogen conditions was found. The mRNA expression level of cyclophilin gene in low nitrogen leaf was increased with the highest of 6.9 folds compared with the mRNA level in normal nitrogen leaf. Gene expression was induced continuously in low nitrogen leaf from 50 days to 70 days the Induced expression of cyclophilin gene by low nitrogen was confirmed by hydroponics experi- ments under control condition which was almost 2.0 folds increased in low nitrogen leaf. Results also showed that leaf growth was mostly affected by low nitrogen which could induce the increase of cyclophilin mRNA expression. Cyclophilin gene might participate in regulating tobacco leaf growth under low nitrogen nutrition stress.
