摘 要:目的建立适应银杏叶提取物总黄酮含量的紫外分光光度测定法。方法比较亚硝酸钠–硝酸铝–氢氧化钠、三氯化铝、盐酸-镁粉、硼酸–柠檬酸、三乙胺5种显色法在350~700 nm波长扫描图谱,分析不同显色方法对银杏叶提取物总黄酮含量测定的适应性。结果三氯化铝显色法在0.0898~0.9878 mg/mL范围内与吸光度(A)线性关系良好,回归方程为y=903.81x+9.6181,r=0.9991,加样回收率为99.3%,RSD=1.5%。样品中总黄酮含量为199.8 mg/g。结论该方法简便、快速、可靠、重复性好,可以适用于银杏叶提取物总黄酮含量测定。Objective To establish a ultraviolet spectrophotometry method for the determination of total flavonoids in extract of Ginkgo biloba leaves. Methods Wavelength-scanning spectra were compared between 350 nm and 700 nm with coloration methods by NaNO2-AI(NO3)3-NaOH, AlCl3, Mg-HC1, boric acid-citric acid and triethylamine, respectively. Adaptability was analyzed for the determination of total flavonoids in extract of Ginkgo biloba leaves with different coloration methods. Results The absorbance and concentration of total flavonoids had a good linear relationship in the range of 0.0898-0.9878 mg/mL. The regression equation was y=903.81x+9.6181, r=0.9991. The average recovery was 99.3 % and the RSD was 1.5%. The total flavonoids content was 199.8mg / g. Conclusion The method can be simple, rapid, reliable and reproducible, and can be used for the determination of total fla'conoids in extract of Ginkgo biloba leaves.
