摘  要:目的探讨3-巯基丙酮酸硫转移酶(3-mercaptopyruvate sulfurtransferase,3-MST)mRNA在正常、急性缺氧及急性缺氧加外源性H2S条件下的表达情况。方法 21只SD大鼠随机等分为正常对照组、急性缺氧组和急性缺氧+NaHS(H2S供体)组。建立大鼠急性缺氧模型,采用反转录聚合酶链反应(RT-PCR)技术,观察各组大鼠延髓3-MST mRNA的表达水平。结果正常大鼠延髓有3-MST mRNA表达;给予急性缺氧刺激后,大鼠延髓3-MST mRNA表达量明显增加(P<0.05);急性缺氧+NaHS组3-MST mRNA表达量比急性缺氧组明显减少(P<0.05)。结论 3-MST mRNA存在于正常成年大鼠延髓组织中,急性缺氧可上调其表达,外源性H2S可抑制由缺氧所引起的3-MST mRNA表达的增加,提示3-MST-H2S途径可能参与急性缺氧大鼠延髓呼吸中枢缺氧损伤保护作用及节律性呼吸的中枢性调节。Objective To investigate the expression of 3-Mercaptopyruvate sulfurtransferase(3-MST)in medulla oblongata of rats under normal, acute hypoxia and acute hypoxia plus exogenous hydrogen sulfide(H2S). Methods 21 Sprage-Dawley rats were randomly divided into normal control,acute hypoxia and acute hypoxia plus sodium hydrosulfide( NaHS)group. Acute hypoxia models of rats were established, then the expression levels of 3- MST mRNA were determined by RT-PCR in the medulla oblongata of each group rats. Results 3-MST mRNA were detected in the medulla oblongata of normal control rats,the expression levels of 3-MST mRNA were significantly increased in the rats with acute hypoxia( P〈0.05 ), whereas the expression levels of 3-MST mRNA were lower in acute hypoxia plus Naris group rats than that of acute hypoxia group(P〈0.05 ). Conclusions The expression of 3- MST mRNA were existed in the medulla oblongata of normal rats. Acute hypoxia can up-regulated the expression of 3-MST mRNA in the medulla oblongata of rats. The increased expression caused by hypoxia-inducible of 3-MST mRNA can be restrained by adding exogenous H2 S. It suggests that H2 S produced under action of 3-MST in medulla oblongata may be involved in the respiratory response and protective effects of respiratory centers injury induced by acute hypoxia.
