抗纤益肺胶囊对肺纤维化大鼠肺组织病理变化及纤维4项的影响  被引量:3

Effect of Capsule of Kang-xian Yi-fei on Pathological Change of Lung Tissue and Four Data of Fibering in Rat Model of Pulmonary Fibrosis

摘  要:目的:观察抗纤益肺胶囊对博莱霉素致大鼠肺纤维化的病理学影响。方法:选用60只健康Wistar大鼠,随机分为6组:正常组、模型组、阳性对照组、抗纤益肺胶囊高剂量组、抗纤益肺胶囊中剂量组、抗纤益肺胶囊低剂量组,每组各10只。正常组一次性气管滴注等量生理盐水,其余各组均采用经气管滴注博来霉素溶液建立肺纤维化大鼠模型。在造模48 h后,各组分别予生理盐水、生理盐水、强的松混悬液、抗纤益肺胶囊高、中、低剂量药,每日1次,共28天。光镜下观察大鼠肺组织病理形态学变化,并测定肺组织羟脯氨酸(HYP),以及纤维4项,即III型胶原(PCⅢ)、IV型胶原(Ⅳ-C)、层粘连蛋白(LN)和透明质酸(HA)的含量。结果:与正常组比较,模型组、阳性对照组HYP明显升高,有统计学意义(P<0.05)。与模型组比较,抗纤益肺胶囊低、中、高剂量组HYP均有明显降低,有统计学意义(P<0.05);PCⅢ、IV-C、LN、HA均明显降低,有统计学意义(P<0.05)。与阳性对照组比较,高剂量组HYP有明显降低,有统计学意义(P<0.05);抗纤益肺胶囊高、中、低剂量组血清层粘连蛋白和透明质酸酶有所降低,但没有统计学意义(P>0.05)。结论:抗纤益肺胶囊抗肺纤维化作用的机制之一,可能是抑制肺组织中纤维4项的表达,以及抑制胶原蛋白合成。Objective : To observe the effect of Capsule of Kang-xian Yi-fei on pathological change, Methods : 60 healthy Wistar rats were divided into six groups randomly, which were normal group, model group, control group, high, middle and low dose treatment group. There were 10 rats in each group. Normal group was given to normal saline with disposable tracheal instillation, and the other five groups were made up into models of pulmona- ry fibrosis with transtracheal instillation of bleomycin. After 48 hours of making up model, they were given to nor- mal saline, normal saline, prednisone suspl. , high, middle and low dose of Capsule of Kang-xian Yi-fei, once a day. At the 28th day, they were killed. The pathological change scope, HYP in lung tissue was determined, and PCIII, IV-C, LN of lung tissue was observed under light micro- and HA were detected. Results:Compared with in normal group, HYP was stepped up in model group and control group, and showed significant difference (P 〈 0. 05 ). Compared with in model group, HYP was cut down in low, middle and high dose of treatment group, and showed significant difference (P 〈 0. 05). And PCIII, IV-C, LN and HA were cut down markedly in, low, mid- dle and high dose of treatment group, and showed significant difference ( P 〈 0. 05 ). Compared with in control group, HYP was cut down markedly in high dose of treatment group showed significant difference ( P 〈 0. 05 ) . LN and HA were cut down in high, middle and low dose of treatment group, but didnl showed significant differ- ence (P 〉 0. 05 ). Conclusion : Capsule of Kang-xian Yi-fei can inhibit the expressions of the four data of fibering and synthesis of collagen protein, which may be one of the mechanisms of action of resisting pulmonary fibrosis.
