摘  要:目的:建立RP-HPLC法测定苦木药材中4-甲氧基-5-羟基铁屎米酮含量。方法:采用Welchrom C18(250 mm×4.6 mm,5μm)色谱柱,以乙腈-0.1%醋酸水溶液(30∶70)为流动相,流速1.0 mL.min-1,紫外检测器检测,检测波长254 nm,柱温30℃。结果:4-甲氧基-5-羟基铁屎米酮浓度在12.0~240.0μg.mL-1(r=0.9999)范围内与峰面积呈良好的线性关系,平均回收率(n=6)为101.2%,RSD为2.3%。结论:该方法简便、准确、灵敏,为2015年版中国药典一部增订苦木含量测定项提供参考。Objective: To develop an HPLC method for the determination content of 4 - methoxy - 5 - hydroxycan- thin -6 -one in Picrasma quassioides (D. Don) Benn.. Methods: Welchrom C18 (250 mm ×4.6 mm, 5 μm) column was adopted and the mobile phase was mixture of acetonitrile and 0. 1% acetic acid(30: 70) with the flow rate of 1.0 mL . min^- 1 ; The detection wavelength was 254 nm, and the column temperature was 30 ℃. Results: The calibration curve was linear in the range of 12.0 - 240.0 μg . mL-1 for 4 - methoxy - 5 - hydroxycanthin - 6- one(r =0. 9999), and the average recovery was 101.2% with RSD of 2. 3%. Conclusions: This method is simple, accurate, sensitive, which can offer references for the quality control of herbal medicines Picrasma quas- sioides ( D. Don) Benn. in China Pharmacopoeia(2015 Edition).
