高低攻击行为大鼠不同脑区γ-氨基丁酸B2受体表达的差异

摘  要:目的通过检测和比较高、低攻击行为大鼠多个脑区内γ-氨基丁酸β2受体(γ-aminobutyricacidreceptorβ2,GABABR2)的表达情况,为探索GABA及其受体在大鼠攻击行为中的作用机制提供线索。方法应用社会隔离加居住入侵法制备高、低攻击行为大鼠模型,分别取正常、高攻击行为、低攻击行为大鼠(n=10/组)的顶区皮质、前额区皮质、下丘脑和海马,运用RT—PCR和Westernblot分别检测3组大鼠各脑区GABABR:mRNA和蛋白表达情况。结果高攻击行为组大鼠顶区、前额区、下丘脑和海马GABABR,RT.PCR/Westernblot电泳条带相对积分光密度值[分别为(0.507±0.049/0.626±0.038)、(0.609±0.049/0.652±0.010)、(0.359±0.030/0.731±0.044)、(0.296±0.054/0.452±0.079)]低于正常组,差异有统计学意义(P〈0.05);低攻击行为组大鼠顶区和海马内GABABR。RT—PCR/Westemblot电泳条带相对积分光密度值均高于正常组,均差异有统计学意义(均P〈0.05),而前额区和下丘脑却低于正常组,差异有统计学意义(均P〈0.05)。结论大鼠顶区、前额区、下丘脑和海马内GABABR2差异表达与高、低攻击行为密切相关,可能是GABA参与调节攻击行为的机制之一。Objective To compare the mRNA and protein expression levels of γaminobutyric acid receptor B2 ( GABABR2 ) in brain regions of male rats with high level aggressive behaviors and low level aggressive behaviors respectively, and provide clues for exploring mechanism of GABA in aggressive behaviors. Methods Wistar male rats were randomly divided into two groups: the normal group and the aggressive behavior group. Then social isolation and resident intruder stresses were used to establish high level and low level aggressive behavior in the aggressive behavior group. The mRNA and protein level of GABABR2 in parietal cortex,prefrontal cortex,hypothalamus and hippocampus of the three groups rats ( n= I0 in each group) were detected using RT-PCR and Western blot respectively. Results The GABABRB: RT-PCR/Western blot relative integrated optical density of parie- tal cortex, prefrontal cortex, hypothalamus and hippocampus in the normal group rats respectively were. Those of the above four brain regions in high aggressive behavior group rats respectively were ( 0. 507±0. 049/0. 626 ± 0. 038 ), (0. 609 ±0. 049/0. 652 ± 0. 010 ), ( 0. 359± 0. 030/0. 731 ± 0. 044 ) and ( 0. 296 ± 0. 054/0. 452 ± 0. 079) were significantly lower (P 〈 0.05 ) compared with the normal group rats. In the low aggressive behavior group rats, the GABABRB2 RT-PCtL/Western blot relative integrated optical density of parietal cortex and hippo- campus increased statistically (P 〈 0.05 ) , while those of prefrontal cortex and hypothalamus decreased obviously (P 〈 0.05 ). all in comparison with the normal group rats. Conclusion Different expression levels of GABABR2 in parietal eortex,preh'ontal cortex,hypothalamus and hippocampus are relative to aggressive behaviors, which might be one of the mechanism for GABA in aggressive behaviors.
