摘  要:向奶牛乳腺上皮细胞中添加β-羟基丁酸钠,利用双向电泳技术进行细胞亚细胞蛋白质组学方面的研究。结果表明,β-羟基丁酸钠对奶牛乳腺上皮细胞亚细胞蛋白表达有显著影响。Imagemaster6.0软件分析出4个表达上调的蛋白点,鉴定分别是ENO1、α-glucosidase、plastin3和ARPC2。通过实时荧光PCR检测以上蛋白在mRNA水平的表达差异,结果均有不同程度的提高,其中以ENO1、α-glucosidase及Arpc2提高显著(P<0.05)。研究所筛选出的蛋白,将有助于进一步揭示奶牛乳腺上皮细胞增殖和泌乳机制。The purpose of this paper was to explore the impact of milk fat synthesis precursors on subcellular proteomics in bovine mammary epithelial cells by adding β-hydroxybutyrate to it, and 2-D electrophoresis technique was used for subcellular proteomics research. Experimental results showed that ,β-hydroxybutyrate sodium had a significant effect on the subcellular proteins in bovine mammary epithelial cells. By the software Imagemaster 6.0, we found that expression of four proteins rised, ENO1, α-glucosidase, plastin3 and ARPC2. Realtime PCR was used to test differentially expressed proteins, expressions rised at different extents, and ENO1, α-glucosidase, ARPC2 rised significantly(P〈0.05). The proteins selected in this experiment would be helpful to further studies on bovine mammary epithelial cells proliferation and lactation.
