益气解毒中药对肝卵圆细胞系WB-F344作用机制的研究

摘　　要：目的研究具有益气解毒功效的牛黄参(牛黄、西洋参)含药血清对大鼠肝卵圆细胞系WB-F344诱导分化及增殖的影响,探讨其作用机制。方法用不同浓度的牛黄参含药血清(5%、10%、20%)作用于WB-F344细胞,以正常大鼠血清(5%、10%、20%)处理的WB-F344为空白组,以单纯培养体系处理的WB-F344为正常组。四甲基偶氮唑盐(MTT)比色法检测细胞的增殖情况,反转录-聚合酶链反应(RT-PCR)检测WB-F344肝干细胞表面标志物甲胎蛋白(AFP)和肝白蛋白(ALB)mRNA表达情况。结果经具有益气解毒功效的牛黄参含药血清干预后,MTT结果显示,10%、20%浓度的牛黄参含药血清能促进WB-F344的增殖(P<0.01),且10%为增殖的最佳浓度梯度;RT-PCR结果显示,经10%牛黄参含药血清处理的WB-F344细胞AFP mRNA的表达随时间的延长逐渐下降(P<0.01),而ALB mRNA的表达则逐渐升高(P<0.01)。结论益气解毒中药可诱导WB-F344细胞向肝细胞特性方向的分化,可促进WB-F344细胞的增殖,这可能是其抗肝损伤、促进肝再生的机制之一。Objective To study the effects of drug-containing serum of Chinese herbs of tonifying qi and detoxication on the rat hepatic oval cell line WB-F344 induced differentiation and proliferation,and explore the mechanism of the method of tonifying qi and detoxication on hepatic stem cells.Methods WB-F344 cells were intervened by drug-containing serum of different concentrations（5%,10%,20%） Niuhuangshen（Calculus bovis and Panax quinquefolius）,with cells intervened by the normal rat serum of different concentrations（5%,10%,20%） as blank control group,and intervened by simple culture system as normal control group.MTT method was used to detect cell proliferation,reverse transcriptase polymerase chain reaction（RT-PCR） was used to detect the mRNA expression of WB-F344 hepatic stem cell surface markers AFP and liver cell surface markers ALB.Results After intervened by drug-containing serum with the function of tonifying qi and detoxication,MTT results showed 10% and 20% concentration of drug-containing serum could promote proliferation of WB-F344（P〈0.01）,10% was optimal concentration for the proliferation.RT-PCR results showed that 10% concentration of drug-containing serum processing decreased WB-F344 AFP mRNA expression gradually（P〈0.01）,while ALB mRNA expression increased gradually（P〈0.01）.Conclusion Chinese herbs of tonifying qi and detoxication could induce the differentiation of WB-F344 cells to liver cell and promote their proliferation,which may be one of the mechanisms of the method of tonifying qi and detoxication in resisting liver damage and promoting liver regeneration.
