摘 要:目的:观察党参、甘草的皂苷和黄酮提取部位对小肠上皮细胞(IEC-6)迁移的影响。方法:以回流提取法分别提取党参和甘草的皂苷、黄酮部位,并用D101大孔吸附树脂对各部位进行分离纯化,以分光光度法检测含量。IEC-6细胞种植于6孔板并培养24小时,划痕法造细胞迁移模型,划痕后加入各种受试药,24小时后观察细胞迁移情况。结果:党参和甘草黄酮含量(以芦丁计)分别为41.73%和50.98%;党参皂苷含量(以人参皂苷Re计)为40.57%;甘草皂苷含量(以甘草酸计)为57.01%。50mg/L的党参黄酮部位和甘草黄酮部位能促进细胞迁移(P<0.05);党参和甘草的皂苷部位对细胞迁移无明显影响。结论:党参、甘草黄酮部位有促进IEC-6细胞迁移作用,党参、甘草皂苷部位对IEC-6细胞迁移无明显影响。To study effects of flavonoids and saponins of Radix Codonopsis and Glycyrrhiza on IEC-6 cell migration. Methods : Extracts containing flavonoids and saponins of Radix Codonopsis and Glycyrrhiza were obtained by refluxing extraction, respectively, and were separa- ted and purified by D101 macroporous resin to obtain flavonoids fraction and saponins fraction contents of the flavonoids and saponins were de- termined by spectrophotometry. Cells were implanted in 6-well plates and cultured for 24h. , then scratched in each well to build cell migra- tion model,and cDMEM contain testing agents was added in twenty-four hours later, the migrating cells were counted to screen the effective fraction for promoting cell migration. Results: The content of flavonoids in the Codonopsis flavonoids fraction was 41.73%, and that in the Glycyrrhiza flavonoids fraction was 50.98% ; the content of saponins in the Codonopsis saponins fraction was 40.57% , and that in the Gly- eyrrhiza saponins fraction was 57.01%. Each of the flavonoids fractions (50rag/L) from radix Codonopsis and Glyeyrrhizae can promote IEC- 6 ceil migration (P 〈 0.05, compared with the control group). The Saponins fractions of Radix Codonopsis and Glyeyrrhizae had no signifi- cant affect on the cell migration. Conclusion: The flavonoids from radix Codonopsis and Glycyrrhizae can promote IEC-6 cell migration, and the Saponins from radix Codonopsis and Glycyrrhizae have no significant affect on the cell migration.
