摘  要:目的:观察黄皮叶乙醇提取物对非酒精性脂肪肝大鼠肝组织ATP、ADP含量的影响,旨在探讨黄皮叶醇提物对肝组织能量代谢的干预作用,为中药防治非酒精性脂肪肝提供实验依据。方法:采用灌饲高脂肪乳剂方法制备非酒精性脂肪肝大鼠模型,用黄皮叶乙醇提取物灌胃给药4周,高效液相色谱法(HPLC)测定观察黄皮叶醇提取对肝组织ATP、ADP含量的影响。结果:ATP回归方程Y=0.138X+0.413,在1.0~100.0μg/ml范围内线性关系良好;ADP回归方程Y=0.283X+1.177,在5.0~160.0μg/ml范围内线性关系良好;与对照组比较,非酒精性脂肪肝模型组大鼠肝组织ATP、ADP含量明显降低(P<0.05);与模型组比较,黄皮叶250~1000mg/kg灌胃给药能提升NAFLD大鼠肝组织ATP、ADP的含量,其中低剂量组(250mg/kg)的ADP含量显著升高(P<0.05),中、高剂量组(500 mg/kg、1000mg/kg)的ATP、ADP含量均显著升高(P<0.05)。结论:本研究建立的检测ATP、ADP含量的HPLC法快速、简便、准确,可为生物组织中能量物质的评价提供依据。黄皮叶醇提取物可提高NAFLD大鼠肝组织ATP、ADP的含量,有效改善NAFLD后肝组织能量代谢障碍,从而保护肝功能。To investigate the effects of ethanol-extractives from Clausena lansium (Lour.) Skeels (wampee peel) on the content of ATP and ADP in hepatic tissue with nonalcoholic fatty liver disease (NAFLD) in rats, which explores the intervention in energy metabolism of liv- er tissue and provides the basis on treatment of NAFLD with traditional Chinese medicine. Methods : The different dosages of ethanol-extrac- tive from wampee peel were intragastric administrated for four weeks on the rat models of NAFLD with feeding fatty milk. An RP-HPLC meth- od was established for determining the content of ATP and ADP in hepatic tissue samples that studied the effects of ethanol-extractives from wampee peel on liver energy metabolism. Results: The linearity range of ATP content was in the range of 1.0μg/ml and 100.0μg/ml (Y = 0.138X + 0.413, r = 0. 9981 ). The linearity range of ADP content was in the range of 5.0μg/ml and 160.0μg/ml (Y = 0. 283X + 1. 177 ,r=0.9996). Compared with blank group rats, the content of ATP and ADP were obviously reduced in hepatic tissue with NAFLD in rats. However, the content of ATP and ADP were significantly increased when the model rats were administrated with 250 - 1000mg/kg - d ethanol-extraetives from wampee peel. Conclusion: The HPLC method for determining the content of ATP and ADP is simple, fast and accu- rate, which can be used for the analysis of energy material in biological organisms. The ethanol-extractives from wampee peel can prominently improve the ATP and ADP content in liver tissue with NAFLD in rats, which can effectively ameliorate energy metabolism disturbance and protect liver function.
